| In recent years, food safety problems of food-borne have become the focus of social problems. New Chinese veterinary medicine feed additives is becoming a hot spot of new veterinary drug research. The Macleaya cordata (Willd.) R.Br. extractive (SA and CHE) has been registered for medical feed additives. But it was not reported in the target animal pig's pharmacokinetic study. This article used the mixed reference substance solution and internal standard solution establishes a method that was rapid,efficient and accurate detection of the four alkaloids of protopine, allocryptopine, sanguinarine, chelerythrine. This work would lay foundation for the target animal pigs pharmacokinetic study and comprehensive development and utilization of the Macleaya cordata.We use the 0.2%formic acid water-acetonitrile for mobile phase. Meanwhile it adopts the gradient elution by Agilent Zorbax SB-C18 gradient elution. The method showed the perfect separation efficiency, peak symmetry and less time consuming. By optimizing the quantitative and qualitative ion pair, fragmentor, collision energy and massspectra condition, that had established a method which possessed high sensitivity and efficiency in accurate detection in the four alkaloids by the liquid chromatography with electrospray ion source tandem mass spectrometry.The method of extraction solvent and pH was optimized in pre-treatment. After preprocessing, the alkaloids ranged from 86.67% to 103.37% were effect by the matrix.LOD between 0.1-0.2 ng/mL, LOQ of 0.5 ng/mL and the concentration of the four Alkaloids has a good linear relationship with peak area in 0.5-100 ng/mL concentration range. In addition, the average recovery was 81%-124% respectively?The (RSD) of intra-day and inter-day precision all are less than 15%and had the good stability under 4?> 25? and -80? at the same time. The methodology index of this method will shed light on the standard that quantitated of the four alkaloids in tissues. |
