Toxocara canis belongs to Toxocara, Toxocaridae, Ascarididea, which infects in small intestine and stomach in dogs and other animals (including human) In order to investigate ectogenesis of Toxocara canis egg, distinguish its infective eggs and grasp the larva migration in chick in vivo, a series of correlational studies were done, which provided a mass of scientific basis for diagnosis, prophylaxis and treatment of canis toxocariasis.In the first part, cox1 genes of 7 Toxocara canis were cloned and sequence analy zed. These cox1 gene sequences are 440bp in length, the intraspecific similarity rates were between 96.7% and 100%, while the interspecific similarity rates were between 86.4% and 90.8%.In the second part, Toxocara canis egg was cultivated using 10 reagent cultural methods in 5 temperature gradients. And Toxocara canis egg was cultivated and in vitro simulating natural environment. The result showed that Toxocara canis egg all could develop in 15℃, 25℃ and 35℃ in both reagent cultural groups and simulative natural environment groups, while incubation developed in each group was invisible in 5℃ and 45℃. In 10 reagent cultural method in vitro, Toxocara canis egg incubation cultivated in 1 mol vitriol developed fastest in 25℃.Incubation developed fastest in fresh water in 25℃ in simulative groups.In the third part, Toxocara canis infected eggs and identification of Toxocara canis larvae migration test in chickens in vivo, it is showed that infective eggs contained larval phase encompassing two rings(font 6, mobile) and larval phase encompassing two rings(font 8,mobile), with the young embryo 238-270μm in length. The result showed that larva was earliest recovered from livers of chicks after 12 hours and lungs after 24 hours.The present study, Toxocara canis eggs incubation in vitro which simulated natural environment was firstly conducted inland, and identification of Toxocara canis infective eggs was firstly made as well. Toxocara canis larva migration test in chick was firstly made inland. The study provided gists for molecular biology, biological characteristics, clinic trial and public healt of Toxocara canis.
