Wheat is the most widely distributed in the world, the planting area is the largest, wheat production directly will affect the country’s food security and the decrease of arable area on a global scale, improving wheat production is imminent. Wheat needs to converted light energy into chemical energy through photosynthesis, ultimately stored in the form of starch and protein in wheat grain, as a result, the quality of photosynthetic efficiency determines the wheat yield. Wheat leaf color mutant is the study of wheat chlorophyll content and photosynthetic efficiency such as the ideal material.In this study, wheat yellow leaf mutant as material created a yellow-green wheat NIL(yellow 1-20 YY, yellow and green 1-20 YG and green 1-20GG), using 168 SSR molecular marker in the distribution of wheat genome and seed gliadin A-PAGE technology, on the basis of genetic background for its identification, photosynthetic characteristics and main agronomic traits were analyzed. Using yellow green 1-20 YG with Shannong1 hybridize, created the 960 strains F2 generations groups, investigated the number of three types of the F2 population YY(yellow) : YG(yellow green) : GG(green), using the distribution on the wheat genome 455 pairs of SSR molecular markers and located on the 2BS 116 to EST-STS primers to detect the F2 population generations. The results show that:1. For the identification of NIL genetic background, on the detection of 168 pairs of molecular markers, only 1 pair of distribution on the 2BS Xcfd238 in NIL 1-20 YY and 1-20 YG, 1-20 GG has polymorphism. The rest of the 167 pairs of molecular markers in 3 NIL have no polymorphism. Seed gliadin electrophoresis of yellow-green NILs are completely consistent, no difference banding. This indicates that the genetic background of yellow and green wheat highly consistent between NILs. After green period leaf color have differences between three strains of NILs significantly, significant difference is found in photosynthetic pigment content, including 1-20 YY for chlorophyll a severe lack of mutants, the photosynthetic performance of the worst, the serious influence its normal growth and development, make its can’t complete the whole growth period and early death;Yellow-green 1-20 YG also belong to the chlorophyll lack of mutants, but its photosynthetic performance affected small, normal growth and development.2. The number of three types of the F2 population that YY(yellow plants): YG(yellow green plants): GG(green plants) 215:498:247(P(2,0.05)=0.51), by χ2 test, the proportions of the separation of F2 generations accord 1:2:1, by Mendel’s law can be concluded that the gene genetic way as incomplete dominant inheritance.3. Use of 455 pairs of SSR molecular markers distributed wheat genome and located on the 2BS 116 to EST- STS primers for 264 strains F2 populations were detected, the results show that a total of 20 pairs of molecular markers, including 8 SSR molecular markers for, Xgpw1148, Xcfd238 respectively, Xwmc25, Xgwm257, Xcfd11, Xbarc124, Xcmwg682, Xgwm614, and Ygl gene genetic distances are 3.39 c M, 3.75 c M, 4.97 c M, 9.16 c M, 18.35 c M, 27.72 c M, 31.77 c M, 35.77 c M; 8 tag EST-STS, respectively BE498358, CJ945085, BF474397, CJ945509, BQ169830, BE591555, BE497251, BF484232, and Ygl gene genetic distances are 6.8c M, 13.38 c M, 13.79 c M, 21.11 c M, 22.77 c M, 39.73 c M, 40.51 c M, 53.52 c M.
