Dairy goat sperm is sensitive to low temperature, although vitality can meet the requirements after thawing, but the pregnancy rate is too low. The cryogenic liquid preservation of dairy goat semen, not only can effectively prolong the storage time but also maintain good vitality, raising the utilization rate of good dairy goats with artificial insemination techniques.In this study, we used Xi-nong Saanen dairy goat as the research object, and studied the cryogenic(4℃) liquid preservation of dairy goat semen. First of all, the semen quality of the dairy goat was detected, a total number of six goats were tested for 10 months. Statistical indicators of semen are as follows: ejaculate volume 1.35±0.54 ml, sperm density25.25±3.35×108/ml, membrane integrity(%) 41.85±3.73, acrosome integrity rate(%)43.10±2.34,sperm motility(%) 71.35±4.28,in accordance with the standard GB20557-2006 of goat fresh semen, which is suitable for semen cryopreservation. Then, the animal origin dilution effect of semen cryopreservation and the influence of vitamin B12 were detected.Diluent is based on Tris- citric acid- fructose- glucose, by adding 15% egg yolk and the base fluid for comparison, results showed that the number of valid days was 6 days, which was significantly higher than the base fluid(p<0.05), membrane integrity could reach 30.91 ±3.27% on 6th days, also significantly higher than the control group(p<0.05). After the addition of vitamin B12, sperm motility was significantly increased, and the 7th day could reach 30.94+5.41(p<0.05). Sperm linear rate(SVL) was 56±4.49μm/s when sperm motility rate was 30%, which was significantly higher than the control group of 34±5.50μm/s(p<0.05).Then improve the cryopreservation process, mainly by reducing the cooling speed, removing seminal plasma through centrifugal, tracing heavy suspension to test whether it is beneficial to extend the preservation time, eventually effective storage time extend from six days to eight days, which is significantly different(p<0.05). Finally tested the effect of semen cryopreservation without animal source diluent and the influence of adding taurine.Results showed that the effective storage time of the diluted solution containing PVA was 4days, and the addition of taurine was prolonged to 5 days. In addition, the sperm membrane integrity, acrosome integrity and MDA content, T-AOC content were compared. There was a significant difference(p<0.05) after the addition of taurine, which showed that taurine had a protective effect on cryogenic liquid storage of dairy goat semen.
