The interspecific hybridization experiment between’yellow sarson’and kale was carried out in this paper. Hybrid offspring was got by the embryo rescue technology, whose authenticity were identified by morphological identification, cytological identification, molecular markers identification and chromosome ploidy detection, the main results were as follows:1.In embryo rescue experiments, the embry ogenes is rate of ovule culture and ovary culture was compared, the influence of the embryo rate of both embryoculture and ovary culture under different culture medium components, specimen timing and darkness treatment was studied. It was found that the embryo rate ofovule culture was higher than that of ovary culture. M1 medium (MS+2 mg/L 6-BA+0.2 mg/L NAA+activated carbon 1.0 mg/L) is the best medium, the best time of thespecimen was 11 days, darkness treatment 24h thereafter lighting treatment has the best culture effect, in which theembryo rate reached 13.55%.2. Morphological identification results of hybrid progeny show that interspecific hybrids between ’Yellow Sarson’ and kale is very different from parent and belong tothe middle type, which are more likely to be female parent in vegetative growth, witthe following characters of tall and erect plant, robust growth, developed main stem, which indicated significant heterosis. Its has green leaf color with hypertrophy and a generous notched leaves, while petiole bears 3-4 on leaflets. As far as reproductive growth concerned, Flower color of female parent is dark yellow, male flower color is close to white, and hybrids is pale yellow.3. Cytology identification results show that its chromosomes number of true hybrid is 38, which is the sum of the chromosome number yellow sarson (2n= 20) and kale (2n= 18).4. Molecular markers identification results hybrids showed that 9 polymorphism SSR markers were screened as follows:scd59-50, scd59-54, scd59-60, scd40-69, scd40-73, scd40-n52, scd40-n49, CB10022,1 SR126. The interspecific hybrids between’Yellow Sarson’and kale were tested with the above 9 polymorphism primers and the results showed that true hybrid has specific bands of both male and female parent.5. Total 11 interspecific hybrids between’Yellow Sarson’ and kale were used for chromosome ploidy detection with parent kale as reference. The ploidyanalysis resu Its show that the peak of the real hybrids plants is 125, which is the sum of parent s the kale and’yellow sarson’.
