In order to explore high efficient, low cost and tuber crops cultivated in open culture conditions, reduce cost of medium, optimization conditions of inoculation and cultivation conditions.1. This paper selects the sodium hypochlorite and Allicin as antibacterial agent and research of sweet potato and potato seedlings, respectively to determine the best optimal concentration of sweet potato and potato, antibacterial agent. The results showed that the sweet potato seedlings in the concentration of sodium hypochlorite is 0.06%, the average gain weight value to 7.7g. The average number of leaves for 20, average root number 8, average shoot number 9. Its average height is 9.5cm. 0.06% sodium hypochlorite as the optimum antibacterial agent of sweet potato seedlings. And potato seedlings in sodium hypochlorite and concentration of allicin were 0.01% and 0.20g/ml average gain weight value for 7.1 g, the average number of leaves for 18, average root number 9, average number of shoots 8. Its average height is 9.5cm. Potato seedling the suitable inhibitive agent is a combination of0.01% concentration of sodium hypochlorite and Allicin concentration 0.20g/ml.2. Simplified the composition of culture medium composition, explores the potato crop growth in carbon source under 1/2. In order to save the cost of culture medium, inhibit bacterial growth under high glucose environment, choose low carbon medium, MS culture medium carbon source by half. The results showed that: sweet potato seedlings and seedlings of potato average increased weight values were 7.4 and 7.6g, mean number of leaves were 18 and 17, average root number 9 and 10, average shoot number 7 and 6, average height of 8.8and 9.6 cm, were not obviously different from those in the control group can used for tissue culture production cycle. The results showed that the contamination rates of sweet potato seedlings and potato seedlings were 16.67% and 13.33%,respectively, and the survival rates were 80% and 83.33%, respectively, and the difference was not significant.4. To optimize culture conditions, by the stable constant temperature incubator culture to put the somaclone in the indoor natural light, the average increase of sweet potato seedlings and seedlings of potato weight values were 6.2 and 5.9 g, the average number of leaves to 12 and 11, the average root number is 6 and 7, mean shoot growth number 4 and 6, the average plant high 7.6 and 8.0cm, and the control group were significantly different.In summary, the sweet potato seedlings best antibacterial agent for 0.06% sodium hypochlorite solution, Potato best antibacterial agent for sodium hypochlorite concentration of0.01% and the concentration of allicin 0.20g/ml,the culture medium was MS culture medium1 / 2 mother liquor and 1 / 2 carbon source, inoculation inoculation room, constant temperature incubator.
