Soil-borne barley yellow mosaic virus disease, which reduces yield and qualities of barley severely, is one of the most important diseases of winter barley in Europe and East Asia. Barley yellow mosaic virus disease is caused by different strains of Barley yellow mosaic virus (BaYMV) and Barley mild mosaic virus (BaMMV) which transmitted by the soil-borne fungus Polymyxa graminis. The most effective approach to control the diffusion of BaYMV and BaMMV is the breeding of resistant cultivar. In this research, six cultivars (Fengnongpi 1, Yunsimai 1, Kongyou 2, Tong 0306, Huadamai 12, Yangnongpi 5) were maked up to complete diallel cross. Investigated the BaYMV grade of the parents, hybrid F1, F2 population in barley yellow mosaic disease nursery of Agricultural College of Yangzhou University. Analysing the inheritance of resistance and mapping the resistant QTL of barley yellow mosaic virus were conducted and the trial were repeated of two years. The main results are as follows:1. Resistance to BaYMV existed significant variance in six parents. The resistance was controlled by inheritance. Disease grade at different investigating period showed significant variance. Even the same cultivar, its disease grade would be various at different investigating period. The analysis should be conducted underlying the disease grade at the most serious period.2. Barley yellow mosaic disease and the main traits such as plant height, internode length below spike, spike number per plant, thousand seeds weight, total weighe were significant negative corelation. This indicated that the disease resulted in the decrease of main traits.3. Analysis of combining ability and regression analysis of Wr-Vr indicated that the degree of barley yellow mosaic disease controlled by additive effect mainly, meanwhile we can not ignored the effect of non-additive. The degree of barley yellow mosaic disease belonged to partial dominance and the inheritance of resistant gene corresponded to additive-dominat model. In six parents, the GCA effect of yangnongpi 5 showed negative and its numerical value was the highest in two years. Yangnongpi 5 was a great material for resistant breeding.4. In 773 SSR primers, there were 240 primers expressed polymorphism between two parents, yangnongpi 5 and huadamai 12. In 240 markers,5 markers (Bmatg0001, GBM1016, GBM1420, HV13GEIII, HvSS1B) showed polymorphism between resistant and susceptible gene pool. The result of single-marker analysis indicated that GBM1016, GBM1420, HV13GEIII were linked to resistance gene and explained 3.6%, 9.3%,5.2% for phenotypic variance explained of resistance to BaYMV respectively. Molecular marker test indicated that the 3H of yangnongpi 5 maybe existed new resistance gene to BaYMV.
