The aim of this study was to explore the different concentrations of melatonin stimulus conditions of Inner Mongolia cashmere hair follicle stem cells Notch signaling pathway in cell differentiation process composition, Notch signaling pathway regulating gene and differentiation of hair follicle cells related reference mark gene (may exist as a potential downstream target genes in the Notch signaling pathway). Initially identified melatonin stimulated under the condition of hair follicle stem cells to differentiate into hair follicle cells in the process of regulation of Notch signaling pathway in the optimal concentration of melatonin and main effecting of receptor, a preliminary study of the stimulation of melatonin Notch signaling pathway related gene between the possible existence of contact and provide certain theoretical basis for research related to Notch signaling pathway.1. Seek to investigate the effect of melatonin on hair follicle stem cell Notch signaling pathway in differentiation process composed gene of Notchl and Hes-1, Notch signaling pathway regulatory gene Numb、Deltex、TGF-β and differentiation of hair follicle cells reference mark gene FGF5、 Versican、 Pax6、 HG (hair growth) expression influence. Through the establishment of the control group (0 mg/ml), 100pg/ml,300pg/ml,500pg/ml,700pg/ml five concentration group on hair follicle stem cells were treated with regular change of liquid culture, and photographed observe cell growth status. Were extracted 0d, 1d,3d,5d,7d,14d and 21 d cell cDNA as template by using real-time quantitative PCR method for detection of nine gene expression and preliminary determination of the stimulation of melatonin on hair follicle stem cells by Notch signaling pathway, differentiation of the optimal concentration is 500pg/ml.2. In the most suitable concentration 500pg/ml were added melatonin receptor antagonist 4-P and antagonist Luz, adding melatonin receptor antagonist and discussing the expression of Notch1、Hes-1、 Numb、 Deltex、 FGF5、 Versican、 TGF-β、-Pax6、HG (hair growth) changes. Try to explore the main antagonist 4-P and Luz in the antagonism of melatonin receptor between the relevant genes may exist mutual contact of melatonin receptor, initially identified the stimulation of melatonin of hair follicle stem cells by Notch signaling pathway on differentiation main effecting receptor is MTLR2, to enrich the Notch signaling pathway of hair follicle stem cells to differentiate into hair follicle cells the genesis and development of molecular mechanism provides a theoretical basis.
