This study aimed to explore the expression quantity of integrin β1, SOX9, β-catenin, BMP2/4, KAP7/8,K5/14, K1/10. melatonin and receptor antagonists in the process of hair follicle stem cells proliferation and differentiation in Inner Mongolia cashmere goat. This study provide a theoretical basis related to hair follicle stem cell proliferation and induce differentiation orientation of Inner Mongolia cashmere goat.1 The study explored the optimal proliferation and differentiation concentration of hair follicle stem cells with the influence of melatonin.The five levels melatonin including 0pg/mL,100pg/mL,300pg/mL.500pg/mL and 700pg/mL were designed to stimulate hair follicle stem cell in Inner Mongolia cashmere goat.we extracted 0d, 1d, 3d,5d,7d,14d and 21d cell cDNA respectively as a template by real-time quantitative PCR integrin β1, SOX9,β-catenin, BMP2/4, KAP7, KAPS, K5/14 and K1/10 eleven genes, according to the results, the melatonin concentration of 500pg/mL was the optimal concentration of cell proliferation, and 500pg/mL were the optimal concentration of differentiation to hair follicle cells,300pg/mL were the optimal concentration of differentiation to epidermic cells. At the same time integrin β1,SOX9, P-catenin, BMP2/4P2/4, KAP7/8, K5/14 and K1/10 have high expression at certain point of time.2 The study explored which cell membrane receptors in hair follicle stem cells differentiate into hair follicle cells, we set up Opg/ml,500pg/mL,500pg/mL+ 4P-ADOT (MT2 antagonists),500pg/mL+Luzindole (MT1+MT2 antagonists) four experimental groups stimulate hair follicle stem cells, we extracted Od, 1d,3d,5d,7d, 14d and 21d cell cDNA respectively as a template by real-time quantitative PCR integrin β1, SOX9, P-catenm, BMP2/4, KAP7, KAP8, K5/14 and K1/10 eleven genes. According to the results,the receptor antagonists group does not appear KAP7, KAPS high expression of the phenomenon, melatonin stimulate hair follicle stem cell differentiation to hair follicle cells via the cell membrane receptors 2.
