Corpus luteum, as a temporary secreting organs in cows, is essential for the development of the early embryos, plant, to maintain the embryo, the pregnancy status of the regulation of early pregnancy has important significance. By luteal protein expression and potential functional analysis, it can reveal the mechanism of the corpus luteum in the cows reproductive physiology process provides a new research perspective. The experiment established by two-dimensional electrophoresis luteal protein expression profile of the spectrum analyzed by PDQuest 8.0 software, differentially expressed proteins were compared between different corpus luteum, and the differences in protein spots were identified by mass spectrometry.By comparing different preparation methods, IPG strips of different specifications, different concentrations of SDS-PAGE gel concentration, optimize test conditions and parameters related to the establishment of cows luteal dimensional electrophoresis protein profiles. Final selection TCA-acetone precipitation samples Method,18cm IPG strips pH4~7 and 12% SDS-PAGE gel concentration to obtain high-quality proteins, isolated proteins 1071 cows spots.By two-dimensional electrophoresis, normal luteal and incidence cyst luteal tissue analysis, analyzes the acquired eight protein spots, Mass successfully identified six: alpha-l-acid-glycoprotein,glial fibrillary acidic protein,nucleophosmin,ubiquitin. found that a-1 acidic protein may be associated with corpus luteum cyst or related tissue caused by inflammation related to glial fibrillary acidic protein may be associated with luteal cyst caused by hormone abnormalities, nuclear phosphoprotein itself not only involved in the ovaries of normal physiological and biochemical processes may also be involved in the cyst corpus luteum, ubiquitin carboxyl-terminal hydrolase may be associated with ovarian yolk and development related to the development.Studied by two-dimensional electrophoresis technique in different periods corpus luteum, Luteal is divided into non-pregnant corpus luteum group, three months and six months pregnancy corpus luteum group, successfully identified six proteins:26s proteasome,Fibrinogen y, Topoisomerase, lamin, Thioredoxin,p-tubulin.26s proteasome regulatory subunit protein may early pregnancy and embryo related activities, fibrinogen y may maintain the role of pregnancy-related, DNA topoisomerase may luteum formation is related with the growth of lamin expression in reducing pregnancy it may be associated with luteal cells activity related to thioredoxin protein.This study was established and optimized cows luteal protein two-dimensional electrophoresis technology system, the technical basis for the differences in the protein luteal proteomics research, but also for the corpus luteum cyst, research and development related to the molecular mechanism of the corpus luteum to provide a reference.
