| Melatonin is a kind of neuroendocrinologic hormone secreted by the mammalian pineal gland. Studies shown that melatonin has involved in the regulation of the complex physiological process like circadian rhythms, seasonal breeding and ovarian function through the membrane receptors. The study found that MT1 and MT2 are expressed in the retina, the pituitary gland, hypothalamus, ovary, testis, gastrointestinal tract and breast. So far, the expression of MT1 and MT2 wasn’t reported in corpus luteum of dairy goats during pregnancy. This study adopts real time quantitative PCR and Western blot to research the protein and mRNA expression of MTR in goats ovary during different pregnant stages, using the immunohistochemical method to research the localization and distribution of MT1 and MT2 in the corpus luteum gland tissue during different stages of pregnant dairy goats. The results of the study are as follows:1. MT1 and MT2 have expression in ovarian tissue during pregnancy. The abundance of transcripts and protein of MT1 and MT2 were significantly greater in earlier and mid-stage compared to later- stage(P<0.01).2. MT1 and MT2 were indeed widely distributed in the corpus luteum gland tissue during different stages of pregnant dairy goats revealed by immunohistochemistry method. The results showed that, MT1 and MT2 have strong immunepositive reaction in entire pregnancy. Especially at the middle stage of pregnancy, almost all corpus luteum cells appeared strong immunepositive reaction. MT1 and MT2 expression have significantly decreased in the late stage.3. RT-qPCR and Western blot results show that MT1 and MT2 protein and mRNA have decreased during the late stage, and significantly lower than the other period(P < 0.01); The results showed the changing regularity of MR protein and mRNA was consistent with MR during the pregnancy.4.Transfected luteal cells deal with MT to find out mRNA and protein levels of MT1 and MT2 were significantly increased; While CCK 8 were used to detect cell activity, and results show deal with MT after 36 h and 48 h the luteal cells activity is significantly increased(P < 0.01). |
