Abstract/Summary:
Secreted phosphoprote in gene(SPP1), leucine aminopeptidase gene(LAP3) and ligand dependent nuclear receptor corepressor gene(LCORL) were selected and studied as growth candidate genes which located on OAR6 36.15~38.56 Mb region in sheep. The whole or part of coding sequence region were cloned; The expression pattern were also tested; SNPs were scaned and its association with growth traits of sheep were anlyzed. The results were as follows:(1)The whole length of coding sequence in SPP1 gene were cloned, and a part length of coding sequence region in LAP3 and LCORL gene were sequenced, and sequece length of 3 genes were 1122 bp, 567 bp and 5413 bp, respectively. The SPP1 gene coding matter probably functioned as a secretory protein involved in the signal transduction, transcription, and may played an important role in the process of transcription regulation. LAP3 gene probably involved in some biological processes such as the signal transduction, stress response, and transcription regulation. The LCORL probably involved in the process of transcription and transcription regulation.(2)SPP1, LAP3 and LCORL genes expressed broadly in rumen, duodenum, hypothalamus, hypophysis, heart, liver, spleen, lung, kidney, muscle, gonad and fat in sheep. The expression of SPP1 gene was the highest in central nervous, and the expression of LAP3 gene was the highest in duodenum, as well as While LCORL gene was the highest in spleen.(3) SNP scanning and its association with growth traits were analyzed by sequencing and PCR-RFLP. The results showed that there was c.132 A>C in SPP1 gene. The correlation analysis between SNP and triats showed that c.132 A>C was significantly correlated with birth weight and yearling weight of sheep(P<0.05), and C allele was the dominant allele. c.187 C>G and c.1116 C>T were found in LAP3 gene, and c.187 C>G was significantly correlated with birth weight, 1-month weight and 3-month weight(P<0.05), and obvious significantly correlated with 2-month weight(P<0.01). c.1116 C>T was evident significantly related to birth weigh(P<0.01) and significantly related to1-month weight and 2- month weight of sheep. At two mutational sites, body weight controled by C allele increased dramatically. The birth weight, 1-month weight and weaning weight controled by CC genotypes were significantly higher than that controled by GG genotype(P<0.05). C was dominant allele in sheep.
