China is a large agriculture country,which consumes plenty of pesticides every year.As known,pesticide is a kind of very useful means of production and it plays an important role in helping crops growing up. Among these pesticides,organophosphorus pesticide is widely used.As one kind of organophosphorus pesticide,profenofos is synthesized in China,and it is used in high quantities.Because of the continuous increasing use of pesticides nowadays,more and more pesticides are released to environment and may have adverse effects to humans.Our work aimes to the possibility of the pesticides to infect the embryo in the pregnant woman’ uterus.The objective setting is:we first made the mouse gestation,then in the continuous four days(E7.5、E8.5、E9.5、E10.5),we gave the pregnant mice three different doses of profenofos by intraperitoneal injection.In the E14,all the pregnant mice were dissect, and the embryos were taken out for evaluating the injury of mouse embryo as follows:(1)To study the affect of profenofos on the morphology of mouse embryo and radial glial cells,we first weighed the embryos and measured the length of the embryos.The results showed that profenofos improved both the weight and length of the mouse embryo,with the dose of 5mg/kg most efficient.In the experiment of paraffin section,we find that profenofos can increase the mitotic figure and apoptosis cell of radial glial cells,and in the stereology computation,we also find profenofos can increase the volume of radial glial cells.(2)To study the affect of profenofos on cholinesterase and antioxidase,we tested the activities of AChE、BChE、SOD、CAT、LDH、GSH in different embryo groups which are treated by profenofos.The results showed that all the different doses of profenofos can decrease the activity of AChE,and the most efficient dose is 5mg/kg.In all treatment groups,the activities of BChE were elevated.Hereinto,the dose of 25mg/kg is the most obvious.The results of the activities of SOD,CAT, LDH,GSH in different mouse embryo groups were 25mg/kg>5mg/kg>control>1mg/kg,control>25mg/kg>1mg/kg>5mg/kg,control>1mg/kg>5mg/kg>25mg/kg,1mg/kg>control>5mg/kg>25mg/kg, respectively.(3)To study the affect of profenofos on the expression of the NTE gene and PON1 gene,we measured the expression of the NTE gene and PON1 gene in different profenofos treatment groups.The results showed that profenofos can promote the expression of NTE gene,the sequence of different group is 1mg/kg>5mg/kg>25mg/kg>control;the next experiment showed that profenofos inhibit the expression of PON1 gene, the result is control>25mg/kg>5mg/kg>1mg/kg.Our work implies us that profenofos has disadvantages to the embryo in the pregnant mouse’ uterus.Though these disadvantages would not affect the survival of embryo,it may be harmful to the development of the offspring’s iNTElligence.As for the people,pregnant women should be away from the pesticide that exit in the environment to keep the infant healthy.
