ObjectivesTo find the best drugs for acute nickel carbonyl poisoning by investigating some enzymes’ activity, cell cycle and the expression of cell cycle related genes of rats with drugs treatment for acute nickel carbonyl poisoning.MethodsThe Sprague-Dawley rats (180-200g) were exposed to nickel carbonyl (250 mg/m3) for 30 min, and 4 and 30 hr later, methylprednisolone, DDC, sodium selenite, shenfuhuiyang injection and methylprednisolone combined with DDC were injected intraperitoneally to them, respectively. Also, we had the exposed group with no drugs administered and the untreated control group.The rats were sacrified respectively on day 3 and 7. Colorimetry was used to detect the activity of enzymes in lung, liver, kidney, heart, and red cells. The cell cycle of hepatocellulars and lymphocyte were detected by FCM, and the expression of Cdc2 mRNA and Weel mRNA in liver tissue were detected by RT-PCR.Results1. The GSH activities in lung and liver, the SOD activities in heart and kidney,and the Na+-K+ATPase, Ca++-Mg++ ATPase in erythrocyte were significant increased in DDC, methylprednisolone and DDC combined with methylprednisolone 4 hr groups.2. Compared with the exposed group, proportion of hepatocellulars at G0/G1 and G2/M phase were reduced in methylprednisolone, DDC, and methylprednisolone combined with DDC treated groups on day 3, and the proportion of G0/G1 and G2/M phase of 30h was higher than that of at 4h. PI of the drug treatment groups increased on day7 compared with the control group, and lower than the exposed group, and which was much lower in methylprednisolone, DDC and methylprednisolone combined with DDC groups.3. The level of Cdc2 mRNA on day 7 was higher while that of Weel mRNA was lower than that of on day 3 in all the treatment groups. Compared with the exposed group, DDC, methylprednisolone and DDC combined with methylprednisolone,4hr groups of sodium selenite and shenfuhuiyang decoction, the expression level of Cdc2 mRNA was higher than the exposed group on day 3 and lower on day 7. The expression of WeelmRNA was less than the exposed group on day 3. Moreover, there were statistical significance between 4hr and 30hr groups of methylprednisolone, DDC, and DDC combined with methylprednisolone.Conclusions1. Methylprednisolone, DDC, or methylprednisolon combined with DDC could decrease the oxidative damage induced by nickel carbonyl in lung, liver,kidney, heart and erythrocyte,of rats and administering drugs to rats at 4hr was superior to that of at 30hr.2. Methylprednisolone, DDC and methylprednisolone combined with DDC had the curative efficacy on the cell cycle arrest of hepatocellulars and lymphocytes of rat induced by nickel carbonyl and administering drugs at 4hr as early as possible was superior to that of at 30hr. There were no obvious effects of Sodium selenite and Shenfuhuiyang Decoction groups on cell cycle arrest.
