Objective::To study the formulation and processing of itraconazole immediately released pellets and investigate the drug release and stability.Itraconazole immediate-release pellets of the structural characterization, quality assessment, a preliminary stability study.Methods:The pellets were coated by Glatt fluid bed technique. Snigle-factor method was used to investigate the several factors afecting the pellets properties (excipients on the drug type and model, the amount of organic solvent) in prescription factors and the technology factors. The drug stability and release were set as observation parameter, on the study of pellets’coating parameter(intake airflow temperature, intake liquid velocity, heat treatment conditions, coating oven jump) to filt, to decide prescription and coating parameter.Results:The results have revealed that the excipients on the drug type and model, different materials were the most critical factors in influencing the physical characteristics of the resulting products; The final preparation technology:HPMC2910(E5) was chosen as the coatingmaterial, the pellets were coated by Glatt fluid bed technique. The final Prescription(1000pills) is make up by blank pill core 200g, itraconazole 100g, HPMC2910 (E5) 150g, polyethylene glycol 20000 18g, dichloromethane 1800ml, ethanol 200ml.Conclusion The method for preparing enteric-coated pellets of itraconazole immediately released pellets was reliable and repeatable.
