Objective To extract the dry roots of Aconitum flavum Hand.-Mazz and gatherdifferent active extracts,study on pharmacological effects of anti-inflammatory andantalgic of the active extract by30%alcohol, and study on chemical compounds of it,provided the basis for exploiting and using Aconitum flavum Hand.-Mazz.Methods We extracted the dry roots of Aconitum flavum Hand.-Mazz.inNingXia with70%alcohol,we got extractum liquidum.Extractum liquidum wasisolated by Macroporous Absorption Resin,with water,30%alcohol,60%alcohol,95%alcohol. Study on pharmacological effects of anti-inflammatory by mice ear-swolleninduced by xylene and egg white-induced toe swollen experiments, study onpharmacological effects of antalgic by mice writhing induced by acetic acid and micehot-plate procedures. We study on chemical compounds of30%alcohol activeextracts by silicagel column,ODS reversed phase column, gel column and TLCisolated methods. We identified monomericcompounds by1H-NMR,13C-NMR,DEPTand HMBC,HMQC,COSY.Results1.We extracted and obtained four different polar active extracts from the dryroots of Aconitum flavum Hand.-Mazz..Water active extracts10.2g,30%alcohol30.15g,60%alcohol8.5g,95%alcohol13.0g.2. In the experiment of mice ear-swollen induced by xylene,30%alcohol activeextract group have significant difference with model group(P<0.05),the inhibition rate is45.17%;in the experiment of egg white-induced toe swollen,30%alcoholactive extract group have great significant difference with model group(P<0.01),theinhibition rate is63.80%. In the experiment of mice writhing induced by acetic acid,30%alcohol active extract group have significant difference with model group(P<0.05),the pain inhibition rate is52.38%. In the experiment of mice hot-plateprocedures,30%alcohol active extract group have significant difference with modelgroup(P<0.05)after30min and45min.3. We all got fourteen monomericcompounds.Now identified seven monomericcompounds.One was carbohydrate:sucrose(compoundⅠ); one was flavonoid: linarin(compoundⅡ);three was steroids: β-sitosterol (compound Ⅲ),.β-daucosterol(compound Ⅵ), β-sitosterol-3,5,6-triol (compound Ⅶ); two monomericcompoundswas alkaloids: aconitine(compound Ⅴ),hypaconitine(compound Ⅳ).Comparedwith literatures, sucrose,linarin,hypaconitine and were first isolated from Aconitumflavum Hand.-Mazz.Conclusion1.30%alcohol active extract have pharmacological effects of anti-inflammatoryand antalgic.2. Seven monomeric compounds isolated from30%alcohol active extract ofAconitum flavum Hand.-Mazz., aconitine was proved have the effects ofanti-inflammatory and antalgic.Other monomeric compounds wether haveanti-inflammatory and antalgic effects should study in next.
