Objective:Both Ranunculaceae Aconitum plants Aconitum tanguticum(Maxim) Stapf and A.naviculare Stapf are the sources of Tibetan Ga list. Its medication site are the whole herb.But in the folk resources surveies, we found that Aconitum phyllostegium Hand.-Mazz is used as Tibetan Ga list by Ba Zhu, who is the director of the Tibetan medicine research institute in Liangshan Yi Autonomous Prefecture. Therefore,Aconitum phyllostegium Hand.-Mazz is included in this study. Tibetan Ga list can invigorate the circulation of blood and relieve pains, which can be also used to treat some fever symptoms and prevent colds. The quality control method of Chinese Pharmacopoeia (1977) and Tibetan Ga list in the Tibetan ministerial and Health Department of the People’s Republic of China drug standards ·Tibetan Book 1 is too simple. This study aims to develop a comprehensive method to control the quality of Tibetan Ga list.Method:Acomplish a herbal textual research on Aconitum tanguticum (Maxim) and Aconitum phyllostegium Hand.-Mazz. We focus on the plant morphology, drug properties and microscopic features of them by the method of pharmacognosy. At the same time, establish a systematic identification of Pharmacognosy for Aconitum tanguticum (Maxim) and Aconitum phyllostegium Hand.-Mazz, including the macro morphology, the micro characteristics of powder and the micro tissue structure of the stem and the leaf. And the establishment of TLC conditions of Aconitum tanguticum (Maxim) and Aconitum phyllostegium Hand.-Mazz was built, which quercetin and kaempferol are the target component. Completed the determination of Aconitum tanguticum (Maxim) and Aconitum phyllostegium Hand.-Mazz, including the moisture content, the total ash content, the acid insoluble ash content and the extract in them. Esbablish the method to determin the content of quercetin and kaempferol in Aconitum tanguticum (Maxim) and Aconitum phyllostegium Hand.-Mazz by high performance liquid chromatography method. At the same time, we accomplish a method for detecting the toxic components in Tibetan Ga list. Take hordenine as reference peak, we establish the chromatographic conditions and standard specific chromatograms of Aconitum tanguticum(Mzxim). Evaluate Aconitum phyllostegium Hand.-Mazz as another group standings of Tibetan Ga list from Pharmacognosy part, TLC and HPLC, We think that the folk medicine is reasonable.Result:Establish a systematic identification of Pharmacognosy for Aconitum tanguticum(Ma.xim) and Aconitum phyllostegium Hand.-Mazz, including the macro morphology, the micro characteristics of powder and the micro tissue structure of the stem and the leaf. Under the same chromatographic conditions, the samples and the target component were in the same position with the same color spots, good separation and moderate Rf value. Completed the determination of Aconitum tanguticum(Maxim) and Aconitum phyllostegium Hand.-Mazz. Esbablish the method to determin the content of quercetin and kaempferol by high performance liquid chromatography method, which the linear regression equation of quercetin was Y= 678145.6355x-13965, r was 0.9996, In the range of 0.076-3.800μg, the linear relationship was good; the linear regression equation of kaempferol was Y= 923106.2053.x+15739, r was 0.99933, In the range of 0.08-4.000μg, the linear relationship was good. The content of the flavonoids in Tibetan Ga list which from different areas were 1.52%-3.39%.The method to detect the toxic ingredients in Tibetan Ga list is established and the result shows that there are no double ester alkaloids in Aconitum tanguticum(Maxim) and Aconitum phyllostegium Hand.-Mazz which is poisonous. It can be used for the quality control of Tibetan Aconitum tanguticum and provide new ideas for the study of Aconitum plants. As hordenine of reference peak, the chromatographic condition of the specific chromatograms of Aconitum tanguticum(Maxxm) was established. The results showed that the specific chromatograms of 8 batches are very similar in the whole,15 common peaks were identified, but the target component of different batches is of great difference. The method is easy to operate. The precision,stability and repeatability were qualified. It can be used for the quality control of Tibetan Aconitum tanguticum and provide new ideas for the study of Aconitum plants. Evaluate Aconitum phyllostegium Hand.-Mazz as another group standings of Tibetan Ga list from Pharmacognosy part,TLC and HPLC, We think that the folk medicine is reasonable.Conclusion:This study establish a comprehensive method for controlling the quality of Tibetan Ga list and provide new ideas for the study of Aconitum plants.
