Objective:To investigate the CXCL12and CXCR4related to therecurrence of adamantimous craniopharyngiomas and analyze thecorrelation between the level of these markers and adamantimouscraniopharyngiomas recurrence.Methods:Twenty Patients who underwent the first time operation bygross total resection in Beijing Tiantan Hospital from June2007to May2009were followed up until May2010. Gross total excisions were achievedby the same group of pediatric neurosurgeons. Patients who underwentradiotherapy or chemotherapy before the operation or only underwentsubtotal or partial resection were excluded. All the samples weredivided into two groups: primary group (11cases) and recurred group(9cases). The definition of primary group is the patient underwenta first-time gross total resection and MRI showed no residual tumor after48h, and there was no evidence of tumor recurrence after one yearfollowed up and did not received any postoperative radio-orchemotherapy. If one-year follow up, the MRI showed clear neoplasm, thepatient was distributed into the recurred group. The expressions ofchanged genes were indentified by immunohistochemistry (IHC) andwestern blot.Results:IHC for CXCL12and CXCR4protein expression showed a cytolemma and cytoblast staining pattern with diverse intensity. The positiveexpression level of CXCL12in recurred group was significantly higherthan primary group (77.8%vs.45.5%, P<0.05), and the positiveexpression level of CXCR4in recurred group was also obviously higherthan the primary group (88.9%vs.63.6%, P<0.05).The level of CXCL12and CXCR4in recurred group was significantly higher than primary groupdetected by western blot. It was found that CXCL12/CXCR4showed morespecificity for distinguishing primary sample from recurred sample.Conclusion:(1)CXCL12and CXCR4were expressed in most ACP,suggesting these foctors involve in the growth regulation process ofthe ACP.(2)CXCL12/CXCR4may be the potential specific bio-marker to predictthe recurrent inclination of ACP, even if the tumor was radicallyremoved.
