Mutant P53protein does not Show onlyloss of the normal tumor suppressor role, but also to promote normal cell Malignanttransformation.Recently, studies have shown that p53and COX-2expression is closely related, yetthe combination of the two studies reported in the literature,The combined detection ofp53and COX-2expression in the prostate cancer, It plays an important role in theundersatanding of process in the development and the diagnosis prognosis of prostatecancer.Objective:By using immunohistochemistry and RT-PCR to detect the expression of COX-2,p53protein in different pathological and clinical staging in prostate cancer tissue andadjacent noncancerous prostate tissue.To discuss the significance of COX-2and p53incarcinogenesis、progression、angiogenesis and their correlations of prostate carcinoma,providing a new approach for the diagnosis and treatment of the tumor.Methods:Firstly,Select between the2004-2009158cases treated by surgery urology patientsin the Guangzhou First Municipal People’s Hospital. The transfer of clinical records andtissue samples, while the establishment of non-tumor tissue adjacent to the tumorcontrol,benign prostate hyperplasia tissue, COX-2immunohistochemistry and RT-PCRdetection, combined with clinical data (age serum PSA level, Gleason score, tumor stage,lymph node metastasis), The statistical analysis was accomplished by SPSS13.0software.Results:Immunohistochemical results showed that COX-2and p53in prostate cancer tissuethan adjacent to the tumor without tumor tissue, benign prostate hyperplasia tissue Highlyexpressed in prostate cancer patients with Gleason score,tumor stage,lymph node metastasis, with statistical significance. COX-2mRNA, p53mRNA expression levels inprostate cancer tissue was significantly higher than the adjacent non-tumor tissue in theprostate carcinoid been verified by RT-PCR to further validate the prostate cancer cellCOX-2mRNA specific expression.Conclusion:Through this research, the COX-2, p53higher expression in prostate cancer, confirmedthe occurrence of COX-2, p53could become a new marker for early diagnosis,judgmentof prognostic and a potential molecular therapeutic target for cancer therapy.. COX-2, p53two kinds of protein expression in PCa tissue, there is a positive combination of detectionfor prostate cancer progression and prognosis prediction to provide a reference value.
