Objective:To study the expression of the human immediate early response 2 gene(IER2)in prostate cancer,with the stage of prostate cancer and the prognosis of patients with prostate cancer,and explore it as a new risk factor for prostate cancer.Methods : The analysis of immunofluorescence quantitative PCR results showed that the relative expression of IER2 in normal prostate epithelial cells was 2.00 ± 0.15,which was significantly lower than that in prostate cancer cells 5.36 ± 0.43(P < 0.05).Detection of IER2 gene expression in prostate cancer cells and human normal prostate epithelial cells by immunofluorescence PCR.The expression of IER2 in prostate cancer tissues and normal tissues was examined by immunohistochemistry.The difference between IER2 expression and age of prostate cancer patients,Gleason score,TNM staging,and tumor were analyzed by SPASS22.0 statistical software and Oncomine database analysis Metastasis.Result : Analysis of the Oncomine database revealed that IER2 mRNA expression was significantly up-regulated in prostate cancer tissues,and that the up-regulation of IER2 mRNA expression was related to tumor metastasis.Immunohistochemical analysis of the prostatetissue chip found that the expression level of IER2 in prostate cancer tissue was significantly higher than that in adjacent prostate tissue(Immunohistochemistry score,Tissue adjacent to carcinoma :6.118±1.947,Tumor tissue:9.135±2.465,P<0.05),and Related to positive lymph nodes and seminal vesicle invasion(P<0.05).There were no significant differences in IER2 positive expression between ages,Gleason score,and TNM staging of prostate tumors.Conclusion:The expression of IER2 was significantly up-regulated in prostate cancer tissues,which was significantly related to lymph node positive and seminal vesicle gland invasion.It is suggested that IER2 may be used as a new risk factor for prostate cancer tumors and it may provide a new plan for early clinical diagnosis and personalized chemotherapy for patients with prostate cancer.
