Objective To investigate the IL-2and IL-10expression in the brain tissue of mice with herpes simplex encephalitis and the possible immunologic mechanism.Methods Mice were randomly divided into4groups with10mice in each:normal control group, HSV-1infected group, Acyclovir treated group, dexamethasone administration group. The mice in last three groups were inoculated with HSV-1solution to make the models of herpes simplex encephalitis. The HSV-1infection group, acyclovir treated group, and dexamethasone treated group were inoculated intracranially with100TCID50(10-2/L) viral solution20uL. Normal control group were infused into culture solution of HSV-120uL. The mice in acyclovir treated group and dexamethasone treated group were fed with ACV (350mg/kg) once a day for6days on the first day after the postinoculation. The mice in dexamethasone treated group were anesthetized with dexamethasone (2mg/kg) on the third day after the postinoculation.The expressions of IL-2and IL-10of the mice brain tissues in each group were compared by using immunohistochemisty. The mice were observed carefully and the score of neurological injury of the mice was assessed. Pathologic slices of brain tissues of mice were used to identify tissue morphology differences. Pearson Correlations was used to describe the correlations of the two cytokines and neurological injury score.Results The neurological injury score was the highest in HSV-1infection group than that in the other groups, and the dexamethasone treated group was the lowest than others (P<0.05). The neurological injury score in acyclovir treated group was between the scores in HSV-1infection group and dexamethasone treated group. After HSV-1infection, IL-2and IL-10was significantly increased compared with that in normal control group (P<0.05). The production of IL-2and IL-10in acyclovir treated group decreased compared with HSV-1infection group (P<0.05), and examethasone treated group decreased significantly compared with that in HSV-1infection group and acyclovir treated group (P<0.05).The correlation of IL-2, IL-10and neurological injury score were all positive correlation(P<0.05).Conclusions The expression of IL-2and IL-10were increased when HSV-1infected the central nervous system. Then both enhanced and suppression immune response co-exist,the clinical of the nerve injury were also apparence. The clinical symptoms was severe, the mortality was mortality.The antiviral drugs inhibited the proliferation of the virus, the expression of IL-2and IL-10were reduced, the immune response and clinical symptoms were also reduce, the survival rate was increased. Dexamethasone treatment in combination with acyclovir can reduce the innune response of the brain, reduce the mortality, and reduce the complications.
