Objective:To explore the therapeutic effects and mechanism of Docetaxel combined with Endostar for PC3cells in vitro. Methods:PC-3cells were randomly divided into four groups:A group (Endostar only), B group (Docetaxel only), C group (Docetaxel plus Endostar), and Control group (no drug), respectively. At the check point, we analyze the proliferation, apoptosis and MMP、MRP、Bcl-2、Bax gene or protein expression of the cells by MTT, FCM,RT-PCR and Western-Blot, respectively.Results:MTT revealed that Docetaxel or Endostar, only or combined, suppressed the PC-3cell proliferation with time dependent manner. After being exposed to Docetaxel plus Endostar for24h,48h and72h, the suppression rates of the PC-3cells were51.0%,68.7%and82.3%, respectively, which were higher than those in A group and B group (P<0.05). Then, FCM showed that G2/M stage ratio of the cells was89.8%in C group, which was significantly higher than that in Control group (P<0.05), also than those in A group and B group, respectively. Furthermore, MMP9, MRP, Bcl-2and Bax mRNA were all exactly expressed in PC-3cells. The mRNA expressions of MMP9, MRP and Bcl-2in C group were significantly lower than those in Control group, A group and B group (P<0.05). Meanwhile, the mRNA expression of Bax in C group was significantly higher than that in Control group, A group and B group (P<0.05). Western Blot showed that the protein expression of Bcl-2and Bax in C group were respectively lower and higher than that in Control group, A group and B group (P<0.05). Conclusion:Docetaxel and Endostar both showed some inhibitory effects on the growth of PC-3cells, with the highest inhibitory effects in Docetaxel plus Endostar group. Its mechanism may be related with the decreased expressions of MMP9, MRP, Bcl-2, and increased expressions of Bax.
