Efficay And Therapeutic Mechanism Of Endostar,A Modified Endostatin,on Malignant Ascites In Mice

Objective:To investigate the effect and mechanism of endostar in therapy malignant ascites in mice.Methods:Mouse models bearing ascites tumors were established via intraperitone al injection Of H22 cell lines. One hundred and twenty ICR mice were randomly divided into 5 groups and received intraperitoneal injection once a day for five (cisplatin)or six(endostar) days:control(0.9%normal saline),endostar group1(8mg/k g),endostar group2(8mg/kg,received injection after H22 cell line injection 24 hour s),combined group (endostar 8mg/kg plus cisplatin 5mg/kg),cisplatin group (5m g/kg),the cisplatin 5mg/kg dose is the total dose and every day every mice only intraperitoneal injection 1mg/kg. Besides the endostar group2,all groups began received i ntraperitoneal injection at the sixth day after mouse model established. Weight,volume of ascites,mindset,life span in each group was recorded respectively.Diarrhea and ot her drug reactions observed?recorded. Detected the levels of Evan's blue dye in the ascites fluid to determine the mouse peritoneal microvascular permeability. Collected mice serum?ascites fluid and the levels of VEGF?bFGF?MMP-2 i n serum?ascites fluid was determined by ELISA assay.Result:1. According to mindset,orexia and whether diarrhea,the endostar group2 mouse livied best, secondly was the endostar group 1.2. Compared with control group,mice weight increased slower in endostar group1? endostar group2?combined group and cisplatin group, among the three treatment group endostar group2 increased slowest.3. Compared with control group, endostar group1?endostar group2?combined group and cisplatin group all could reduce the peritoneal microvascular permeability of mouse(P<0.05),the volume of peritoneal ascites and prolong the life span, moreover the combined group is most obviously.4. Compared with control group, endostar group1?endostar group2?combined group and cisplatin group all could decrease levels of VEGF?bFGF?MMP-2 in serum and ascetic fluid (P<0.05),and the combined group is most obviously.Conlusion:endostar combined with cisplatin could improve the efficacy because of its more significantly decreasing the levels of VEGF?MMP-2 in serum and ascites fluid.
