Objective: probing into the expressions and significance of HIF-1a and ET-1incolorectal cancer,provide the basis for the point therapy of colorectal tumor.Method: draw on diagnostic norm and exclusion norm, selected50cases(male38cases, female12cases) of colorectal cancer who were confirmed by pathologicaldiagnosis from yanan university affiliated hospital during June2012-September2013.And,selected para-carcinoma tissue specimens of40cases (24cases of male, female16cases),30cases of normal specimens,(male15cases, female15cases),using theimmunohistochemical SABC method to detect the positive expression rate of HIF-1aand ET-1above3groups.Collecting and sorting data to statistic analysis of data bySPASS16.0software, analyzed the relationship with pathological features and theirrelationshipsResults:1.The expression about HIF-1a, ET-1in above three groupsthe positive expression rate of HIF-1a and ET-1in colorectal cancer was96.0%(48/50) and92.0%(46/50), both in para-carcinoma tissue was70.0%(28/40) and70.0%(28/40), both in normal specimens were16.7%and20%. HIF-1a, ET-1in the tissue ofcarcinoma positive expression rate is significantly higher than its expression in thepara-carcinoma tissue (x2=12.664, P <0.05; x2=12.270, P <0.05). Both expressedsignificant differences in the two teams (P <0.05); HIF-1a, ET-1in the tissue ofcarcinoma positive expression rate is significantly higher than its expression in normalcolon tissue (x2=53.998, P <0.01; x2=43.635, P <0.01).Both expressed significantdifferences in the two teams (P <0.05); HIF-1a, ET-1in the tissue of para-carcinoma tissue positive expression rate of is significantly higher than itsexpression in normal colon tissue (x2=20.559, P <0.01; x2=20.559, P <0.01).Bothexpressed in both teams were remarkable different(P <0.05); And HIF-1a, ET-1bothin colorectal tumor team and adjacent team appear proportional relationship(P <0.05).2. HIF-1a, ET-1in the expression and clinical pathological parameters of colorectalcancerHIF-1a is majorly expressed in the plasma of cancer cells, small part in the cellcenter. The positive expression rate relate to colorectal cancer invasion, lymph nodemetastases,and Dukes stage (X2=4.427, P <0.05). The positive expression rate withDukes stage from A to D gradually increased, having lymph metastasis is remarkablemore than without lymph metastasis (76.5%vs33.3%); HIF-1a express has nothing todo with the tumor size and degree (P>0.05).ET-1is remarkable expressed in cancer cell center.The positive expression rate andpositive strength are higher than normal colon mucosa tissues, and relate to tumor attack,lymphatic metastasis and Dukes staging, with lymph metastasis had a remarkable stronger positive expression rate and positive strength without lymph metastasis group (73.5.%vs50.0%).3. the expression of HIF-1a and ET-1with gender and ageAccording to the gender group, the explicit expression rate of HIF-1a, ET-1incolorectal cancer were97.4%(37/38),94.7%(36/38) in men patients, women in patientswith colorectal cancer were91.7%(11/12),83.3.%(10/12).Examined by the X2methodsand gender teams had no remarkable difference between the two teams (P>0.05);According to patients’ age group, with60for community,>60colorectal cancer patientswith positive expression rate of100.0%,89.2%respectively, the <60patients withcolorectal cancer in the positive expression rate is90.9%,95.5%, have examined by theX2method and age groups had no remarkable difference during the both(P>0.05)4. The correlation of HIF-1a,and ET-1in colorectal cancer In50cases of tumor samples, HIF-1a,and ET-1at the same time expressed aspositive rate was83.7%(36/43) respectively, both expressed negative for4.6%(2/43).Collecting related data using Spearman rank correlation analysis, we discoverythat HIF-1a, ET-1correlation expression and there was a positive correlation betweenrelations(X2=6.319),correlation coefficient is0.315, P <0.001.Conclusion: HIF-1a, ET-1has strong expression in colorectal tumor, particularin having distant metastasis and lymph node metastases and in the para-carcinoma tissueand normal tissue showed lower expression or without expression, And both expressionin colorectal tumor was positively correlation. HIF-1a expression and co-ET-1canhave a pivotal role in the mechanism of tumor blood formation and attack,Therefore,itmay provide important targets for clinical target therapy for colorectal cancer.
