Objective: By using the animal model of typical rat SAH, to discuss the expressionpattern and intervention meaning of correlation factor Bcl-2、Bax, and the apoptosis,vasospasm of cereborvascular endothelial cell after SAH.Methods: To make the SAH model using the twice-injection of blood, dividing60SD ratsof SAH into three big groups stochasticly,they are group A(group SAH)、groupB(groupSAH and group NS) and group C(group SAH and group of cyclosporine intervention).Then dividing three groups into five subgroup(each group has4rats), using TUNNELmark method to detect the apoptotic positive cell of blood vessel endothelium in differenttime point after killing animal at the corresponding time point. Usingimmunohistochemistry (IHC) to observe the apoptosis of correlation factor Bcl-2、Bax ineach group, and using Imge-proplus(Ipp) to detect the IOD value of blood vessel.Results:1, TUNET detection finds that vascular endothelial cell and apoptotic cell ofsmooth muscle are beginning to appear after3hour of SAH, and apoptotic cell isincreasing,vascular endothelial cell(VEC) is beginning to appear at1d. Apoptotic cell isincreasing obviously then getting a climax value after5d of SAH, both apoptotic cell andpositive cell of ciclosporin intervenegroup are decreasing in different time point, especiallyobvious at3d and5d;2,expression of protein of Bcl-2in group A and group B isdecreasing gradually along with the time extension of SAH, beginning to go down at3d,the IOD getting a minimum value at5d, positive cell of Bcl-2in group C is beginningto increase from SAH to3d, the IOD getting a climax value at5d, it is significant tocompare the difference between group A,group B and group C(P﹤0.01);3,positive cell ofBAX in group A and group B is increasing obviously at3d after SAH, both the enhance ofexpression intensity, the IOD getting a climax value at5d, the expression oh Bax isdecreasing with the time extension of SAH, it is meaningful to compare the differencebetween group A,group B and group C. Conclusion:1, endothelial cell of blood vessel of brain and the phenomenon of smoothmuscle along with CVS after SAH;2,Cyclosporine A could remit the apoptotic endothelialcell of blood vessel of brain, and smooth muscle to some extent after SAH, and could havecertain therapeutic of cerebral angiospasm after SAH.
