Cynanchum otophyllum Schneid (Asclepiadaceae), named asQingyangshen, which is widely distributed in the southwest of China. It is afolk medicinal plant endemic to the southwest of China, and its roots wereused for the treatment of kidney weakness and rheumatic pain by the localpeople. Recently, in our research, the chloroform extract of Qingyangshenshowed apparent antidepressant activity in mice, and the C21steroidalglycosides were found to be the active ingredients. In this thesis, the chemical constituents of the cultivated Cynanchumotophyllum were studied. With the combined application of differentchromatography methods, sixteen compounds were isolated, and theirstructures were elucidated by spectroscopic and chemical methods, especiallytwo-dimensional NMR techniques. The structures of these compounds wereidentified to be qingyangshengenin-3-O-β-D-glucopyranosyl-(1â†'4)-β-D-cymaropyranosyl-(1â†'4)-β-D-oleandropyranosyl-(1â†'4)-β-D-cymaropyranosy-l-(1â†'4)-β-D-cymaropyranoside (1), qingyangshengenin-3-O-β-D-glucopyranosyl-(1â†'4)-β-D-glucopyranosyl-(1â†'4)-β-D-oleandropyranosyl-(1â†'4)-β-D-cymaropyranosyl-(1â†'4)-α-L-cymaropyranosyl-(1â†'4)-β-D-cymaro-pyranoside (2), caudatin-3-O-β-D-glucopyranosyl-(1â†'4)-β-D-cymaropyranosyl-(1â†'4)-β-D-cymaropyranoside (3), caudatin3-O-β-D-glucopyranosyl-(1â†'4)-β-D-thevetopyranosyl-(1â†'4)-β-D-cymaropyranosyl-(1â†'4)-β-D-digitoxopyranoside (4), caudatin-3-O-β-D-glucopyranosyl-(1â†'4)-β-D-thevetopyranosyl-(1â†'4)-β-D-cymaropyranosyl-(1â†'4)-β-D-cym-aropyranoside (5), caudatin-3-O-β-D-glucopyranosyl-(1â†'4)-oleandropyranosyl-(1â†'4)-β-D-cymaropyranosyl-(1â†'4)-β-D-digitoxopyranos-ide (6), caudatin-3-O-β-D-glucopyranosyl-(1â†'4)-β-D-cymaropyranosyl-(1â†'4)-oleandropyranosyl-(1â†'4)-β-D-cymaropyranosyl-(1â†'4)-β-D-digitoxopy-ranoside (7), lostamin-3-O-β-D-glucopyranosyl-(1â†'4)-β-D-glucopyranosyl -(1â†'4)-β-D-cymaropyranosyl-(1â†'4)-oleandropyranosyl-(1â†'4)-β-D-cymaro-pyranoside (8), qingyangshengenin-3-O-β-D-glucopyranosyl-(1â†'4)-β-D-oleandropyranosyl-(1â†'4)-β-D-cymaropyranosyl-(1â†'4)-β-D-digitoxopyranos-ide (9), qingyangshengenin-3-O-β-D-glucopyranosyl-(1â†'4)-β-D-glucopyranosyl-(1â†'4)-β-D-cymaropyranosyl-(1â†'4)-oleandropyranosyl-(1â†'4)-β-D-digitoxopyranoside (10), qingyangshengenin-3-O-β-D-glucopyranosyl-(1â†'4)-β-D-glucopyranosyl-(1â†'4)-oleandropyranosyl-(1â†'4)-β-D-cymaropyranosyl-(1â†'4)-β-D-cymaropyranoside (11),qingyangshengenin-3-O-β-D-glucopyranosyl-(1â†'4)-β-D-cymaropyranosyl-(1â†'4)-oleandropyranosyl-(1â†'4)-β-D-cymaropyranosyl-(1â†'4)-β-D-digitoxopy-ranoside (12), caudatin-3-O-β-D-glucopyranosyl-(1â†'4)-β-D-cymaropyranosyl-(1â†'4)-oleandropyranosyl-(1â†'4)-β-D-cymaropyranoside(13), caudatin-3-O-β-D-glucopyranosyl-(1â†'4)-β-D-glucopyranosyl-(1â†'4)-β-D-cymaropyranosyl-(1â†'4)-oleandropyranosyl-(1â†'4)-β-D-cymaropyranoside(14), caudatin-3-O-β-D-glucopyranosyl-(1â†'4)-β-D-glucopyranosyl-(1â†'4)-β-D-cymaropyranosyl-(1â†'4)-oleandropyranosyl-(1â†'4)-β-D-digitoxopyranosi-de (15), caudatin-3-O-β-D-glucopyranosyl-(1â†'4)-β-D-cymaropyranosyl-(1â†'4)-oleandropyranosyl-(1â†'4)-β-D-cymaropyranosyl-(1â†'4)-β-D-cymarop-yranoside(16). Among them, compounds1-8were new compounds, andnamed as cynanotophylloside A-H.
