Objective: In this study, different concentrations of Chinese herbal medicine QilanGranulates were used to act on human tonguel squamous cell carcinoma cells Tca8113.By investigating the effects of Qilan Granulates on inhibiting proliferation and inducingapoptosis of those cells in vitro, we mainly provide an experimental foundation forclinical application. Methods: Tca8113cells and human gingival fibroblast cells arerandomized into blank control,6.25mg/ml、12.5mg/ml、25mg/ml、50mg/ml、100mg/mlgroups, respectively cultivated for24h:1. The morphological changes were observed bythe inverted phase contrast microscope.2. The growth inhibition rate of Tca8113wasassayed by CCK-8.3. The apoptosis of Tca8113was analyzed by flow cytometry.4. Theexpression of Bax protein and Bcl-2protein in Tca8113were analyzed by Western blot.Results:1. The results of cell morphology indicated that Tca8113grew border and clearin blank control group while the Tca8113what were treated with different concentrationsof Qilan Granulates presented characteristic morphological changes of apoptosis.The cellmorphology of human gingival fibroblast cells did not change much.2. CCK-8resultsshowed that the proliferation of Tca8113was inhibited significantly by differentconcentrations of Qilan Granulates in a does-dependent manner.3. The results of flowcytometry showed that the apoptosis rate of Tca8113was increased with the increasing ofQilan Granulates concentration.4. The results of WB showed that the expression of Bcl-2protein was significantly down-regulated after treated with Qilan Granulates for24hours,while the expression of Bax protein was up-regulated. Conclusion:1. Qilan Granulatescould inhibit proliferation and induce apoptosis of Tca8113.2. Down-regulation of Bcl-2protein expression and up-regulation of Bax protein expression might be one of the mainmechanisms for apoptosis of Tca8113.
