Objective:To carried out a preliminary study on Chinese traditional medicine-gout1, and toprovide experimental evidence for treating gout.Methods:Mice hyperuricemic model were made by xanthine600mg/kg ig, potassiumoxonate200mg/kg ip. The blood uric acid level was detected by automaticbiochemical analyzer. And XOD level in the liver was detected by enzyme labeling.Use the method of HE dye to observe histopathologic changes on mice kidney. Ratgouty arthritis model was made by uric acid sodium solution, and observe jointswelling after dosing. Use ELISA to detect the levels of TNF-α and IL-1β, observepathology changes on the joint.Results:The mice was dosed after7days, the blood uric acid level of model increasedfrom (87±22.3)μmol/L to (145±33.4)μmol/L, the XOD level increased from(41.40±1.25)U/L to (50.48±1.21)U/L, they had significant difference from thecontrol group(P<0.05). In the result of histopathologic observation, it showed thatglomerular structure was normal, the size of tubule was same, and inflammatory cellswere not be seen in the Kidney, it hadn’t caused any damage in mice kidney byxanthine ig, potassium oxonate ip, and it showed that the hyperuricemia model wassuccessful.21days later,the blood uric acid level decreased from (138±30.2)μmol/Lto (98±26.1) μmol/L, the XOD level decreased from (53.62±3.24)U/L to(42.45±2.42)U/L, they had significant differentce from the model group(P<0.05). But the uric acid level hadn’t significant difference from the allopurinol group andbenzbromarone group(P>0.05), it showed that the gout1had the same effect withallopurinol and ben-zbromarone.2. In the rats gouty arthritis model, the jointswelling was very obvious at48h, it increased from (1.88±0.12)ml to(2.16±0.13)ml, and it had significant difference from the control group(P<0.05). HistopathoIogicexamination showed that the synovial cells were seriously injured in the model group.The TNF-α level increase from(241.43±35.56)pg/ml to(346.98±22.65)pg/ml, andIL-1β level increase from(142.54±30.88)pg/ml to(184.68±34.42)pg/ml,they hadsignificant differences from the control group(P<0.05),it showed that the model wassuccessful. After treating,the gout1could significantly inhibit joint swelling of rats,the joint swelling decrease to (1.90±0.16)ml,it had significant difference from themodel group(P<0.05). And the gout1reduce the levels of TNF-α and IL-1β andmarkedly improve the histopathologic changes.Conclusion:The gout1(plantago, gentian, clematis)can decrease the blood uric acid ofhyperuricemic mice what’s made by xanthine ig, potassium oxonate ig. Themechanism for gout1may be relevant in inhibiting the activities of XOD. In additionto, the gout1possessed antigout arthritis effects,and it’s mechanism may be relatedto inhibit the product of inflammatory cytokine (TNF-α and IL-1β). Chinesetradional medicine-gout1may become a new drug for curing gout.
