Background:HSP90is a class of widely present in eukaryotic cells highly conservedmolecular chaperones, a growing number of studies have shown that it plays animportant role in tumor cell proliferation and differentiation, survival, and mobile andangiogenesis, has become a new cancer treatment target.According to whether theycontain rich snippets of glutamic acid amine, Human HSP90divided into twocategories, HSP90ɑ and HSP90β, recent studies have shown that HSP90ɑ, not only inthe cell and the expression on the cell surface, even secretion to work outside the cell,and is closely related to high tumor invasion and metastasis.Primary hepatocellular carcinoma is one of malignant tumors with highmorbidity and high mortality, has become a worldwide problem of serious harm tohuman health, molecular biology and epidemiological studies have confirmed thatHepatitis B virus (HBV) infection is a major cause of chronic hepatocellularcarcinoma (HCC).Because of HCC mortality is extremely tall, its mainly in the high mortality ratewas found to have to middle-late, cancer cell metastasis and recurrence after surgeryhas also led to the high mortality rate. Therefore, early detection, early diagnosis and early treatment and effective treatmentis a key to improve the5-year survival rate of liver cancer patientsDetection of serum markers of liver cancer is one of the main means of earlydiagnosis, at present, the AFP (Alpha fetoprotein, AFP) of primary liver cancer is stilla clinical serological markers, but its sensitivity, specificity and accuracy are not ideal,especially for cancer patients with liver cancer, less than3cm in diameter sensitivitywas33%-65%;If the AFP levels greater than20ng/mL for diagnostic standard, itspositive rate to fall further to9.1%-26%.Currently found serum marker is moreassociated with liver cancer, but there’s no more superior than the AFP tumor-specificmarkers, independently and effectively to the early diagnosis of liver cancer and todetermine the prognosis.Therefore, urgent need for new and more accurate and reliable markers of livercancer in order to improve the sensitivity and specificity of early diagnosis of livercancer.Studies have shown that, HSP90α over expressed in HCC tissues, but HSP90αand early diagnosis of HBV-related HCC invasion and metastasis rarely carried out.To this end, we detect hepatocellular carcinoma cell lines with different metastaticpotentials and HBV-related hepatocellular carcinoma in patients with serum HSP90αexpression levels, analyzed its expression with clinicopathological parameters ofrelevance to explore with HCC diagnosis, invasion, metastasis and prognosisrelationships.Objective:1)Detection of heat shock protein90α(HSP90α) expression levels in differentmetastatic potential of human hepatoma cell lines and HBV-related hepatocellularcarcinoma (HCC) patients in serum;2) To explore the clinical significance of abnormal expression HSP90α.Methods:1) Using Western Blot technique to detect the expression levels of HSP90α in different metastatic potential of hepatocellular carcinoma cell lines (no metastaticpotential HepG2cell lines, high metastatic potential MHCC97H cell lines).2) Using ELISA technique to detect the patients with HBV-related hepatocellularcarcinoma, benign liver disease patients and healthy controls serum HSP90αexpression levels in serum; retrospective analysis of clinical data of patients withHBV-related hepatocellular carcinoma, single factor analysis of the relationshipbetween the expression of HSP90α and clinicopathological data of HCC cases.Results:1)HSP90ɑ are expressed in the two kinds of cell lines,the expression level of HSP90αin high metastasis potential MHCC97H cells was significantly higher than thatwithout metastasis potential HepG2cells (P=0.0002).2)HBV related hepatocellular carcinoma patients and patients with benign liverdiseases HSP90ɑ expression level in serum were significantly higher than that ofhealthy controls (P=0.0001),but the difference between HBV-related hepatocellularcarcinoma and benign liver disease was not statistically significant (P=0.990).HBV-related hepatocellular carcinoma in patients with serum HSP90α expressionlevels correlated with age, HBeAg is positive, AFP levels, HBV DNA levels andTNM stage were not significantly correlated with sex and related (p=0.044).Conclusions:1). HSP90α associated with the invasion and metastasis of HBV-related HCC.2). HSP90αis expected to become one of the invasion and metastasis of HCC andprognosis assessment and distinguish HBV-related liver disease and healthy markers.
