Objective The preparation technology of1α-hydroxydehydroepiandrosterone,which wasstudied, was obtained for industrialized production.Methods Starting from androsta-1,4-diene-3,17-dione,1α-hydroxydehydroepiandrostero-ne can be obtained through eight steps such as protection of carbonyl group,bromination,elimination, epoxidation, reduction of carbonyl group, hydride reduction, ring-opening anddeprotection. Several steps were optimized. The structures of taget compound and someintermediates were comfimed by MS,1H-NMR,13C-NMR.Results Getting the taget compound, the preparation technology of1α-hydroxydehy-droepiandrosterone was complished,and a part of the preparation technology was optimiz-ed. The total yield is13.7%. The structures of taget compound and some intermediateshad been established by MS,1H-NMR,13C-NMR.Conclusions The preparation technology of1α-hydroxydehydroepiandrosterone whichhad been obtianed, has easily available raw materials, conventional reagents with all ones, mildreaction conditions, simple operation, and lays a foundation for industrialized production. |
