Objective:To investigate the effect of stellate ganglion block on multiple organ functionand the serum levels of TNF-α and HMGB1in sepsis in ratsMethods:The sepsis rats established by cecal ligation and puncture(CLP),the stellateganglion block rats established by transection of cervical sympathetic trunk(TCST).Seventy-two healthy male SD rats,weighing250-300g, were randomly divided into3groups (n=24each): Sham group (group S), Cecal ligation and puncture group(groupCLP),Cecal ligation and puncture group+Transection of cervical sympathetic trunkgroup(group CLP+TCST).At6h,12h,24h and48h after the start of experiment,bloodsamples of each group were collected to detect serum necrosis factor-α (TNF-α) andhigh mobility group box1(HMGB1) levels.At the same time,aspartate transaminase(AST),alanine aminotransferase(ALT), creatinine(Cr), blood urea nitrogen(BUN),creatine kinase(CK),creatine kinase-MB(CK-MB)levels were detected byautomatic biochemical analyzer to assess the function of liver, kidney,heart.Thenthese rats were killed at each time point and their heart, lung, liver and kidney tissueswere kept for histological examination.Result:1、The activity of CLP rats changed significantly,performance for hair erected,curled up, drank less, shortness of breath, the serious symptom performance for eyes,nasal exudate appeared.Compared with the CLP group,the symptoms of CLP+TCSTgroup rats were lighter,most performance for curled up, drank less.S group rats actedfreely.2、Compared with group S,the TNF-α levels at each time points of CLP groupwere significantly increased(P<0.01),the TNF-α levels at each time points of CLP +TCST group were increased(P<0.05),and both peaked in the6h point; Comparedwith group CLP, the TNF-α levels of CLP+TCST group were significantlydecreased(P<0.01).3、Compared with group S, the serum HMGB1levels of CLP group started toincrease at12h (P <0.05), and peaked in the24h point(P <0.01),the serum HMGB1levels of CLP+TCST group started to increase at24h point (P <0.05);compared withthe CLP group, the serum HMGB1evels of the CLP+TCST group at each time pointwere decreased.In the24h,48h point,the difference was significant (P <0.01).4、Compared with group S, the ALT,AST,Cr levels of CLP group started toincrease at12h point,BUN increased at6h point (P<0.05),and all reached the peak at24h point(P<0.01).CK, CK-MB began to increase at6h point(P<0.05),and peaked at12h(P<0.01); compared with the group S, the serum ALT,AST,Cr,BUN,CK, CK-MBlevels of CLP+TCST group both increased at12h,24h time point(P <0.05); comparedwith the CLP group,the serum AST,ALT,BUN,Cr,CK,CK-MB levels of CLP+TCSTgroup at each time point were decreased(P<0.05), in the24h point,the AST,BUN, Crlevels were significantly decreased (P<0.01).5、Microscope observation the morphology of heart, liver, lung, kidney tissueshowed: the tissue morphology of S group were normal;the tissue morphology ofCLP group were severely damaged, cell disorder, widespread edema, necrosis andinflammatory cell infiltration. Compare with the CLP group, the tissue morphology ofCLP+TCST group changed slightly with mild edema and cell spotty necrosis.Conclusion:stellate ganglion block can depress the serum levels of TNF-α and HMGB1insepsis in rats, and improve the functional and histological indicators of multipleorgan,paly a role of multiple organ protection in CLP rats.
