Objective: To discuss the application of Tip-spray ionization massspectrometry(Tip-spray MS) in the diagnosis of esophageal cancer and Try to findmetabolic differences of medium and small molecular substances in esophagealcancer tissue and normal esophageal tissue.Methods:Using Tip-spray MS with parameters as follows: positive ion mode,mass range50-1000Da, atomization voltage3.5kV,spraying distance2cm,lensvoltage65.0V,ion transmission tube temperature150℃,to analyze the tissue samplesfrom22esophageal cancer patients and the matching tissue adjacent to carcinoma andprincipal component analysis(PCA) to process dates from the Mass spectrumfingerprint spectra.Results: Tip-spray MS can catch stable esophageal tissue signals in onesecond.Peptide peaks at m/z values of104,154,175,611,655,699,743,787and831showed significantly different relative abundance. Peaks at m/z values of611,655and699, which corresponded to porphyrin, had the most obvious difference. PCAcould effectively distinguish esophageal cancer and tumor-adjacent normal tissue.Conclusion:1.Tip-spray MS combined PCAcan identify esophageal cancer andnormal tissue adjacent to carcinoma effectively, which may contribute to the rapiddiagnosis of esophageal cancer.2.As the material basis for the diagnosis of esophageal cancer, thedifferent metabolism of porphyrin molecular may play an important role in theoccurrence and development of esophageal cancer.
