Analysis of animal tissue samples is of great importance in the field of medicine,food safety,etc.but most of the traditional analytical methods require complex pretreatment processes,such as grinding,extraction,separation,etc.The sample pretreatment is often time-consuming and lengthy.More seriously,some valuable material information within the tissue samples may get lost so result in analysis is relatively low.Therefore,using a green,fast and non-destructive detecting technique is very important.Internal extractive electrospray ionization mass spectrometry(iEESI-MS)is one of the new types of ambient mass spectrometry technology.The most unique feature of iEESI-MS is that it can obtain the chemical information of the internal components of complex solid samples without any sample pretreatment.The method is simple operation,high speed of analysis,high sensitivity and high throughput.This paper focuses on the application of iEESI-MS technology in lung cancer tissue samples for amino acid and meat quality appraisal.The main contents of this paper includes as follows:1.The analysis of amino acids in lung cancer tissue and adjacent tissue samples using iEESI-MS.1)Application of lung cancer tissues and adjacent tissues,that iEESI-MS technology can directly and quickly identify the chemical composition of amino acids,qualitative and quantitative analysis,without any pretreatment sampling,without gas assistance.And PCA was applied to distinguish the differentiation lung cancer tissue and adjacent tissue samples with a little bit of data of iEESI-MS fingerprints.Providing a powerful technical support for the rapid detection of clinical tissue samples 2)The comparison of iEESI-MS with conventional DESI-MS mass spectrometry was conducted,the iEESI could obtain more abundant chemical mass spectrometry information from both the interior and surface samples with higher sensitivity and lower detection limits.The results showed that the iEESI-MS has both qualitative and quantitative ability in animal tissue samples analysis with simple operation,the high speed of analysis.2.iEESI-MS was applied for the study of illegal additives in meat tissue samples.The results of the study on the pork,mutton,beef and supermarkets beef samples from Jiangxi institute of sports science show that the iEESI-MS method can detect and identify trace “clenbuterol” substances in meat,establishing the mass spectrometry platform for direct analysis the “clenbuterol” component of meat.3.Analysis of trace meat quality using iEESI-MS.The fingerprints of trace lean pork and trace fat pork tissue samples were obtained by iEESI-MS and the nutrient components in the meat were identified via collision-induced dissociation(CID).Principal component analysis(PCA)was applied to better visualize the differences for mass spectral fingerprints of different kinds of pork.Our experimental results indicate that iEESI-MS allows rapid quality recognition of complex matrix animal tissue samples within rich material information.Which can provide a reliable detection tool for the analysis and research of nutrient in food,and also has important practical significance in meat safety detection.
