Objective: To evaluate the quality of the Edible Bird’s Nest by SDS-PAGE, HPAE-PAD, HPLC, Kjeldahl method and ICP-MS and establish the quality standard for the Edible Bird’s Nest. It may provide scientific foundations to the authenticity identification, quality control and the further development and utilization of the Edible Bird’s Nest.Methods:1. The protein of Edible Bird’s Nest and adulterants were identified by SDS-PAGE.2. Precolumn Derivation HPLC was used to determinate the sialic acid in the Edible Bird’s Nest and adulterants.3. HPAE-PAD was used to determinate the sialic acid in the Edible Bird’s Nest and adulterants.4. Kjeldahl method was used to determinate the protein in the Edible Bird’s Nest and adulterants.5. Ion chromatography method was used to determinate the nitrite and nitrate in the Edible Bird’s Nest and adulterants.6. ICP-MS was used to determinate the heavy metal content in the edible bird’s nest and adulterants.Results:1. The SDS-PAGE method was established to identity the Edible Bird’s Nest protein and it can be effectively used to qualitative identification.2. Precolumn derivation HPLC method was established to determinate the sialic acid in the Edible Bird’s Nest, the method is simple, accurate and reliable.3. HPAE-PAD method was established to determinate the sialic acid in the Edible Bird’s Nest, which is less material and time-consuming, without derivative, low toxicity, accurate and reliable.4. Ion exchange chromatographic method was established to determinate the nitrite and nitrate in the Edible Bird’s Nest, the method is simple, accurate and reliable. Couclusion:1. SDS-PAGE can be accurately used to identify the authenticity of the Edible Bird’s Nest, can be used for adulteration identification of protein-containing material is doped, but not accurate for the merits of the pseudo-doped and quality identification.2. Sialic acid in the Edible Bird’s Nest is in the form of N-acetyl neuraminic acid,it has physiologically active, and content is the higher, and thus can be used as index components nests.3. Pre-column derivatization HPLC and HPAE-PAD can be accurately used to determinate the sialic acid in the Edible Bird’s Nest, which may be provide scientific foundations to the authenticity identification.Compared with the Pre-column derivatization HPLC, the HPAE-PAD is more simple operation, need less time, do not need derivative, need less consumables and low toxicity.4. The Edible Bird’s Nestt is asialoglycoprotein composition which is full of amino groups, It may produce carcinogens-nitrite under certain conditions of humidity and temperature,and the environment containing nitrates. The experiment conditions have a great influence on the result.
