Eucommia ulmoides is a valuable Chinese herbal medicine resource in China, whichis a second nationally-protected plant. Its fruits, roots, leave and flowers have very highmedicinal values. They contain rich natural active substance and the alkaloid part hasstronger activity than other parts. Previous studies are mainly focused on its flowers andthe extraction of the effective components from it, but its leaves and alkaloids areseldom studied. The alkaloids were extracted through acid soluble alkaloid precipitation,organic solvent extraction, macroporous resin and silica gel column chromatography,and thin layer chromatography. Then the pharmacological activity of the extraction wasinvestigated using activity screening method. The results provide the basis for theeffective use of resources of Eucommia ulmoides.(1) In this experiment Eucommia ulmoides leaves were taken as the object ofresearch. At the same time, the classical acid soluble alkaline extraction was used toisolate and purify total alkaloids. This study was conducted on the basis of single factor,and by response surface analysis, the optimal extraction parameters were obtained. Theresult of the analysis showed that the optimum extraction conditions of total alkaloidswere the hydrochloric acid pH value of2, ethanol concentration69%, ratio of materialto liquid1:17.(2) By using the macroporous resin purification method, the total alkaloids ofEucommia ulmoides were purified, thus removing impurities. The experimental resultsshowed that the largest amount of alkaloid could be obtained on condition that themacroporous adsorption resin type was NKA-9, Ethanol concentration was55%, theconcentration of sample was0.02g/ml, the pH of sample liquid was9, and theconcentration of sample was0.03g/ml. The total alkaloid of Eucommia ulmoides wasisolated by silica gel column. In our study we used thin layer plate as eluant. Theexperimental results showed that: the best elution ratio of chloroform, ethyl acetate andmethanol was38:38:24. At last,4alkaloids were purified; each of them was named sample1,2,3and4respectively.(3) This research is about the antioxidant and antibacterial activity of the totalalkaloids separation and sample1,2,3,4. The activity of the purified extractions wassignificantly higher than total alkaloids. The alkaloids from eucommia leaves showeddifferent levels of inhibition for5various kinds of bacteria (Staphylococcus aureus,Escherichia coli, Bacillus subtilis, Aspergillus, Penicillium). In general, the purifiedalkaloids could better curb the growth of these bacteria than the total alkloids. Inaddition, the results of total reducing capacity determination, DPPH radical scavengingactivity experiment and anti-lipid peroxidation ability determination indicated thatthere was no significant difference between ascorbic acid and the purified alkloidsextracted from the total alkaloids in terms of their antioxidant capacity.
