With the wide application of Eucommia ulmoides, studies focused on chemical composition of Eucommia ulmoides received more and more attentions by researchers. Herein, the active ingredient in Eucommia ulmoides were investigated generally. This paper consists3works as follow:1. Multi-step extraction procedures with organic solvents were used towards Eucommia ulmoides extract followed by applying column chromatography to separate the collected chloroform layers. Factors such as specification and amounts of silica gel, choice of eluent were discussed. A pure product was isolated after the sample was treated twice with column chromatography. It was characterized by UV and NMR, and compared with previously published works. A conclusion that the product is SDG can be made. The purity of SDG is94.37%and the overall yield is0.58%.2. Established a reliable method by using high-peRformance liquid chromatography to determine3kinds of biologically active components simultaneously. The conditions of extraction and chromatography have been investigated, the UV wavelength at210nm,240nm,280nm and320nm were used to detect aucubin, geniposidic acid, chlorogenic acid and pinoresinol diglucosid simultaneously. This method was demonstrated to be accurate with linearity, repeatability, reliability.3. Rutin was chosen as standard substance to determine the amounts of flavonoids from4different extraction methods and reflux extraction is preferable. The optimal conditions were:the solid-liquid ratio was1:12(g/mL), the concentration of is60%, the extraction times is2and1.5hours for once. Flavonoids in Eucommia ulmoides, which were obtained from different locations, were determined and analysed by HPLC. The PCA results showed that Eucommia ulmoides from different locations has different amounts of flavonoids, and this method can be applied for identifying of Eucommia ulmoides true or false.
