Objective The dissertation is to disscuss the possible drug-resistance mechanismsof gliomas based on the studies on the expression of O6-methylguanineDNAmethyltransferase (MGMT), nuclear factor—kappa b (NF–κB) and GlutathioneS-transferases-π (GST-π) in gliomas and the correlation among the three genes in theirexpression in gliomas.Methods All the seventy specimens were obtained from the glioma resectiondone by the Nerve Tumor Surgical Department of General Hospital of Armed PoliceForces in Beijing from July2010to July2011. The expression of protein wasinvestigated in70cases of malignant glioma specimens of the human brain and20cases of MGMT, NF–κB and GST-π in normal brain tissue by immunohistochemistry.The correlation among MGMT, NF–κB and GST-π and the relation between themalignancy, age of onset and gender and the gliomas were analyzed.Results The positive expression rate of MGMT protein, GST-π protein and NF-κB p65B in normal brain tissue and gliomas is different. The difference is ofsignificant meaning (P<0.05).The positive expression rate of MGMT protein in high and low-grade gliomagroup is different. The difference is of no significant meaning (P>0.05). The positiveexpression rate of GST--π protein and NF–κB p65B protein in high and low-gradeglioma group is different. The difference is of significant meaning (P<0.05).The positive expression rate of MGMT protein, GST-π protein and NF-κB p65Bprotein is different in onset age of glioma and gender. The difference is of significantmeaning (P<0.05).The three drug-resistance genes MGMT, NF-κB and GST-π have no remarkablerelation (P>0.05).Conclusion MGMT, NF-κB and GST-π in the malignant gliomas of the human brain have a higher expression which is unrelated to the gender and age of the patient.The expression of MGMT in gliomas is unrelated to the severity of gliomas. Theexpression of NF-κB and GST-π is significantly related to the severity of gliomas. Thethree drug-resistance genes MGMT, NF-κB and GST-π have no remarkable relation.There are phenomena of co-expression of multiple drug-resistance genes in gliomacells. The inspection of the expression of multiple drug-resistance genes at the sametime is instructive to drug use in individual chemotherapy of the patient after gliomasurgery.
