Objective:Establish animal model of oral mucosal reconstruction with nanostructuers silkFibroin matrices and acelluar dermal matrix.Evaluation the feasibility of nanostructuerssilk fibroin matrices as mucosa repair scaffold in the present study. By comparing wtithAcelluar dermal matrix with was widely using in clinical to determine the effectiveness ofthe silk scaffold in clinical applications.Methods:1.Animal experiments80male Wister rats, randomly divided into four groups.Full thickness defect(10mm indiameter)on the buccal mucosa were created by surgical incision with a standardblade and under microscopic guidanc.Then20rats were randomly assigned per group.Group A: repairing oral buccal defects with nanostructure silk fibroin (10mm in diameter),n=20;Group B repairing oral buccal mucosal with cowhide cell-free matrix(10mm indiameter), n=20, C Group repariing with human skin acellular(10mmin diameter) indiameter, n=20; Group D as blank control group, the buccocal mucosal defects werecovered with vaseline gauze (10mm indiameter), n=20;General and immunohistochemicalobservation after1week,2week,3week,4week.2. Histological observation of the reconstruction oral mucosa:1).Grawn the oral mucosa in postoperative1week,2weeks,3weeks,4weeks from thewhole layer of materials of the oral mucosa.2).General observation then after HE staining and immunohistochemical staining,Gynamic evaluation the changes of the new newborn oral mucosa. Result:1.The rats model of reconstructiong oforal mucosa was successful.all the surgical incisionswere healed as scheduled.No postoperative wound infection was found in all group,and noconstracture or out of the materials was seen.The growth of the regeneration oral mucosawas good.one week after surgery.2.The reparing effect of the nanometer fiber silk fibroin and the cell matrix material fororal mucosa is almost the same,but the time of initial epithelium is Slightly later.One weekafter surgery the silk fibroin material neatly arranged,the inflammatory reaction aroundthe material is obvious, there were more inflammatory cells, fibroblasts and a smallamount of blood capillary, haven’t seen a epithelial structures. Two.week..aftersurgery,newborn epithelial cells can been seen around the surgical wound edge,the newlayer is thinner, layer number of3-4,arrangement of the epithelial is irregular, epithelialcell growth active,small blood vessels can been seen in Lamina propria,collagenarrangement of lamina propria was loose.inflammatory reaction was mitigation.thenumber of inflammatory cell decrease fibroblasts and capillary number increases.silkfibroin material partly degradation and absorption.three week after surgery,newbornepithelial become thicker, which was multilayer structure,there were more epithelial footand nail,Multilayer structure of the epithelial become poloidal,which arranged moreclosely, inflammatory cells in the lamina propria significantly decrease,but more cleariyblood vessels.Collagen arrangement in the lamina propria more close from the previous.Silk fibroin scaffold degradation into pieces.Four week after, the newborn epithelial wasmore thicker,which was multilayer structure, epithelial shackles processes, there wsa noobvious difference between normal oral mucosa of rats.Collagen arrangement in the laminapropria is neat.Keratin epithelial spectrum antibody immunohistochemical study prove thatthere was epithelial tissue in the newborn oral mucosa.Conclusion:1.To furtherly improve the animal experiment,prove that the wister rat is the idealexperimental animal in disease research. 2.From the observation of short-term around erioperative and postoperative,prove thefeasibility of using nanometer fiber silk fibroin materials to repair of the oral mucosa, andbecause its production materials is from natural silky,which is wide variety of sources,lowprices, so there are broad application prospects insilk fibroin materia.3. Nanometer fiber silk fibroin materials can promote the oral mucosa epithelialhyperplasia, accelerate wound healing, inhibiting wound contraction of the oral mucosa,reduce scar contracture.
