Objective To observe the influences of low molecular weight heparin (LMWH) on the expressions of hepatocyte growth factor(HGF),vascular endothelial growth factor(VEGF) in the lung tissue of and interleukin-6(IL-6) in the plasma of rats with chronic obstructive pulmonary disease(COPD),then the interventional mechanism for COPD of LMWH would be demonstrated.Methods Thirty Wistar rats were randomized to be divided to three groups:①normal control group:rats were kept in normal condition,with no intervene;(2)COPD model group:The COPD rats model was established by smoke inhalations and endotracheal instillations of lipopolysaccharide,normal saline was injected subcutaneously as control;③LMWH intervened group:models were set up as above, low molecular weight heparin (150U/kg,about1mL)were injected subcutaneous half hour before smoke.Lung tissue histopathology change,blood gas analysis and the total and differential cell counts of bronchial alveolar lavage fluid(BALF)were determined. HGF and VEGF in lung tissue were detected by immunohistochemistry,and IL-6in serum was detected by ELISA.Correlation analysis were carried out between HGF,VEGF, IL-6and the numbers of neutrophils respectively.Results The lung tissue histopathology changes of COPD model group were coincident with that of humans,the rat lung tissue pathology change in LMWH intervened group reduced compared with model group, the quantitative analysis have significant difference(P<0.05);blood gas analysis showed that the PaO2and PaCO2were different in the three gropus significantly(P<0.01),and so as the total weight blood cells and neutrophils of BALF and HGF and VEGF in lung tissue and IL-6in serum.in LMWH intervened group,the PaO2were elevated and the PaCO2droped compared with COPD model group(P<0.05);the total weight blood cells and neutrophils of BALF and HGF and VEGF in lung tissue and IL-6in serum in LMWH intervened group were decreased compared with COPD model group,the difference had statistical significance(P<0.05).The relationship between HGF,VEGF, IL-6and the numbers of neutrophils were positively correlated(P<0.01).Conclusion HGF,VEGF and IL-6were all involved in the airway inflammation of COPD,LMWH may decrease the expressions of them to alleviate the inflammation process.
