Nasal polyps(nasal polyps,NP)is a common disease,frequently-occurring disease,the reported incidence of 1% to 4%,accompanied by nasal,nasal sinus mucosal chronic inflammatory changes,due to its complex and complex postoperative recurrence of the characteristics and therefore in the ENT disease plays an important role.The cause of nasal polyps is unknown.Cytokines are of concern in many causes of nasal polyps,whether it is a high degree of edema of the nasal mucosa,submucosal stroma and vascular proliferation,or glandular secretion and lamellar infiltration of lamina propria Are associated with more or less cytokines.The expression of interleukin 25(IL-25),interleukin 13(IL-13)and vascular endothelial growth factor(VEGF)in nasal polyps were detected by immunohistochemical SP method.Analysis of the relevance of the three,and then to explore the role of cytokines in the process of nasal polyps play a role.Objective:In this study,we investigated the expression of IL-25,IL-13 and VEGF in nasal polyps and normal nasolabial mucosa tissues by studying the expression of IL-25,IL-13 and VEGF in the formation of nasal polyps The role of the three and whether there is a certain interaction between,so as to a more comprehensive understanding of the pathogenesis of nasal polyps,to further explore the etiology of nasal polyps and conservative treatment provides a molecular basis and new ideas.Materials and methods:The experiment was divided into three groups: nasal polyps group,chronic sinusitis ethmoid sinus mucosa group and normal inferior turbinate mucosa control group.Chronic rhinosinusitis(with nasal polyps),chronic rhinosinusitis(not associated with nasal polyps),and nasal septum deviation from endoscopic surgery in the first hospital of Baoding City from February2014 to June 2015 The patients with the inferior turbinate hypertrophy were used as experimental materials.30 cases of nasal polyps were from chronic rhinosinusitis(associated with nasal polyps)patients: 16 males and 14 females,aged 18 to 65 years;ethmoid sinus mucosa specimens from 30 cases of chronic rhinosinusitis(not accompanied by Nasal polyps)patients: 16 males and 14 females,aged 18 to 60 years;normal nasal mucosa tissue specimens from 10 cases were from the nasal septum deviation associated with turbinate hypertrophy patients: 6 males and 4 females;age 18 ~ 55 years old.All patients were excluded from allergic rhinitis,bronchial asthma,aspirin poorly tolerated and other systems of serious illness.All tissue specimens were fixed by 4% formaldehyde,alcohol dehydrated,impregnated with wax,tissue sections.The expression of IL-25,IL-13 and VEGF in the above samples was detected by immunohistochemical SP method.SPSS19.0 statistical software was used to analyze the data.Results:1 H&E staining Light microscopic display: nasal polyps tissue,ethmoid sinus mucosal tissue eosinophil infiltration increased significantly,the distribution is limited to mucosal subcutaneous blood vessels and glands around.And eosinophil infiltration in the normal nasal mucosa slightly seen.There was significant difference between the two groups(P <0.01).2 The expression of IL-25 in nasal polyps,ethmoid sinus mucosa and inferior turbinate tissues IL-25 was expressed in nasal polyps,was strongly positive,positive expression in ethmoid sinus mucosa,mainly in expression rates of IL-25 in nasal polyps group,ethmoid sinus mucosa group and inferior turbinate group were 83.3%,73.3% and 10%respectively,and there was significant difference between the two was weakly positive.The positive expression rates of IL-13 in nasal polyps group,ethmoid sinus mucosa group and inferior turbinate group were 90.0%,80.0%and 10% respectively,and there was significant difference between the two groups(P <0.01).4 The expression of VEGF in nasal polyps,ethmoid sinus mucosa and inferior turbinate tissues VEGF in the nasal polyps and ethmoid sinus mucosa mainly located in the epithelial cells,glands and blood vessels and vascular walls around the endothelial cells,the basement membrane around the inflammatory cells are also expressed,mostly located in the cytoplasm,a few located in the cell nuclear.VEGF is weakly expressed in normal nasal cavity.The positive expression rates of VEGF in nasal polyps group,ethmoid sinus mucosa group and inferior turbinate group were 86.7%,76.7% and 20% respectively,and there was significant difference between the two groups(P <0.01).5 Correlative analysis of IL-25,IL-13 and VEGF expression in nasal polyps Correlation analysis showed that: there was a positive correlation between IL-25 and IL-13 expression in nasal polyps(r = 0.400,P <0.01),IL-13 was positively correlated with VEGF expression(r = 0.632,P <0.01)VEGF expression was also positively correlated(r = 0.738,P <0.01).Conclusion:1 One of the important pathologic features of nasal polyps is the increase in eosinophil infiltration.2 IL-25 can induce Th2 immune response,enhance the proinflammatory response,promote the secretion of Th2 cytokines and cascade amplification of inflammatory response,induced tissue secretion of inflammatory mediators to Eos aggregation and epithelial cell proliferation,resulting in nasal polyps.3 IL-13 can activate Th2 and inhibit Th1 immune response,regulate T,B cell function,promote Ig E production increased,causing inflammatory cell infiltration,induced Eos wandering,involved in nasal polyps.4 VEGF can induce tissue to produce a large number of microvessels,to speed up the growth of nasal polyps,followed by neovascular plasma extravasation,nasal and paranasal tissue accumulation,tissue edema,local compression caused by epithelial cells hypoxia,rupture,nasal polyps gradually formed.5 IL-25,IL-13 and VEGF were positively correlated with the expression of polyp in the polyps,suggesting that the three factors may be involved in the process of nasal polyps,and confirmed that the development of nasal polyps is the result of multi-factor cross-effects.
