| Objective To investigate the progress of intimal thickening and the expression of toll-like receptor4(TLR4) after vascular balloon injury in rats, and to discuss the effects of them on restenosis and the effects of valsartan on them.Methods Rats were randomly allocated into three groups:control group (n=20), operation group (n=22) and valsartan group (n=22). The operation group and valsartan group were given aortic endothelial denudation, and the valsartan group were given valsartan(20mg/kg/d) from1day before injury to14and28days after aortic injury. The control group were given sham operation. The progress of intimal thickening was detected by HE stain. The expression of TLR4mRNA and the level of protein of TLR4were investigated at day14and28after injury by reverse transcription polymerse chain reaction(RT-PCR) technique and immunohistochemistry method, respectively.Results The proliferation of vascular smooth muscle cells(VSMC) and the intimal thickening were observed at14days and28days after balloon injury. Compared with the control group, the expression of TLR4mRNA and protein increased obviously in the operation group, and they all decreased at28days than at14days. When given valsartan, the intimal thickening and the expression of TLR4decreased apparently.Conclusion1. The expression of TLR4increase obviously after vascular balloon injury in rats;2. Valsartan inhibit the expression of TLR4, and prevent intimal thickening. |
