Objective:Using a certain concentration of TNF-a, IL-6to interfere the fetal rat osteoblasts in vitro, to detect the cell activity and the expression of its cells’perlecan mRNA.Methods:Extract and subculture fetal rat calvaria osteoblasts through enzymatic digestion method, with a certain concentration of TNF-a, IL-6to interfere its growth, CCK-8method was used to detect the osteoblast proliferation activity in different periods, while using RT-PCR method to detect the osteoblasts perlecan mRNA expression.Results:At24h,48h,72h, compared with the control group, the experimental group (TNF-a group, IL-6group) significantly reduced the activity of osteoblasts, perlecan mRNA expression was significantly lower (P<0.05).Conclusion:In cultured fetal rat osteoblasts, a certain concentration of TNF-a, IL-6can reduce the activity of its cells and inhibit their expression of perlecan mRNA.
