Objective:This experiment use the allogeneic bone marrow mesenchymal stem cells to cure the wound in rats.And to observe the efficiency and safety of bone marrow mesenchymal stem cells injection for wound healing.Methods:(1)The rats of wound divide into groups;(2)The rats bone marrow mesenchymal stem cells of rats isolation, culture, purification, amplification and batches;(3)Using flow cytometry instrument identificate the immune phenotype of rats bone marrow mesenchymal stem cells;(4)Use BrdU staining marks the rats-bone marrow mesenchymal stem cells;(5)Local injecte the the rats-bone marrow mesenchymal stem cells in the rats;(6)Analyze the difference of wound healing rate between the experimental group and control group in the statistical analysis.(7)Using Western blot to detect the express of ikeratinocyte growth factor in two groups.(8)Histopathological observation of the wound.Results:(1)We successful isolate and obtain the rats bone marrow mesenchymal stem cells. The positive expression rates of cell phenotypes were various as followings respectively: CD29(99.99%), CD105(99.22%), CD44(100%)and the negative expression rates of cell phenotypes were:CD45(0.50%)respectively. It is demonstrated that a pure rats bone marrow mesenchymal stem cells has been obtained.(2)From the appearance of wound healing, the experimental group is better than the control group.Two groups of wound healing rate also significantly statistical differences.(3)According to the staining grading standards of the healing skin after HE.he experimental group is better than the control group.(4)According to the result of Western Blot.the KGF protein expression of he experimental group is higher the one of the control group.Conclusions:Using the allogeneic bone marrow mesenchymal stem cells may be an effective and safe method for the wound in rats.
