BackgroundBreast cancer is the most common malignant tumors of women, its incidence is increasing at every year2%-8%speed, has now become the first killer of women’s health. Triple negative breast cancer refers to estrogen receptors and progesterone receptors and human epidermal growth factor receptor2were negative for a breast cancer subtypes, With poor tumor differentiation, strong invasion and postoperative prone to recurrence and metastasis features. The BRCA1gene is the most common breast cancer susceptibility gene, Many studies show that breast cancer BRCA1genetic correlation and triple negative breast cancer is not only similar in immunohistochemical phenotype but also gene expression is also very close, At present, the study of BRCA1gene and the prognosis of triple negative breast cancer has not been carried out.ObjectiveIn this study, through multiple SNP sites of BRCA1gene were genotyped, research of each genotype and three negative breast cancer in relation to prognosis.Methods280cases triple negative breast cancer patients treated in Cancer Hospital, Chinese Academy of Medical Sciences and Peking Union Medical College in recent five years were taken into the study, with complete demographic,clinical and pathological data, Patients were divided into two groups according to whether they had recurrence or metastasis. According to international human gene group monomer type figure plans (http://hapmap.ncbi.nlm.nih.gov/) provides of SNP public database to retrieve the BRCA1gene minor allele frequency (MAF)>0.05of SNP bit points, select qualified15SNP sites, Sequenom’s Mass ARRAY system was used to detect the genotypes of each SNP of BRCA1gene in the group of patients, further analysis of the relationship between the genotypes of the SNP and three negative breast cancer prognosis. Result1, The study concluded relapse transfer group patients and disease free survival was not significantly different in patients with age, family history of breast cancer, pathological type, histological grading (P>0.05). But there was a statistically significant difference in clinical stage, whether the vascular invasion, tumor size and lymph node metastasis (P<0.05);2, The study concluded that the genotypes of15SNPs of BRCA1gene in recurrence group and disease free survival between group frequencies are quite, and there is no obvious statistical difference (P>0.05). Conclusion1,Late clinical stage, vascular invasion, large tumor size and lymph node metastasis were important factors in triple-negative breast cancer patients with poor prognosis.2,There is no significant correlation between BRCA1gene polymorphism and the prognosis of TNBC.
