ObjectiveOn the basis of establishing animal model of selectivity damaged SGNs to investigate the protective effect of Brain-derived neurotrophic factor(BDNF) on the Spiral ganglion neurons(SGNs) treated with ouabain. In order to provide the basis for clinical work to find the drugs of protecting SGNs,we need to study the change in cell morphology and density, the main role of BDNF in the cochlea site and the impact on organelles.MethodsThe round window membranes of Mongolian gerbil were instilled with ouabain (80umol/L) to establish animal model of selectivity damaged SGNs, then the treated round window membrane was instilled with BDNF (5.4ug/ml).Removed the cochlea after7days, prepared specimens for electron microscope, which was examined by transmission electron microscope. Histopathological changes were observed and analyzed by staining with HE, the density of SGNs was calculated. We could explore the main role of BDNF in the cochlea site, and the impact on organelles,then semi-quantitative analysis of SGNs density where measured by Image-pro plus6.0.All the dates were analysised by SPSS17.0, Differences associated with P values of <0.05were considered as statistically significant.ResultsThe protection effect of BDNF on SGNs focused onthe base of the cochlea,the density of R1and R2were higher than others, The difference was significant compared with the ouabain groups (P<0.05).SGNs of the Mongolian gerbil was damaged when ouabain was instilled on theirs round window membranes, the density of SGNs was decreased, the morphous changed greater, organelles was damaged especially mitochondria. The difference was significant compared with the control groups (P<0.05).BDNF could rival the ototoxicity of ouabain in the cochlea, the protection was more obvious.It could be confirmed about SGNs that the morphous changed smaller,the density was higher. The difference was significant compared with the ouabain groups (P<0.05)ConclusionsA certain degree of protection of BDNF instilled on the round window membranes of Mongolian gerbil. The main role of BDNF in the cochlea site could be the base of the cochlea, that was the R1and R2.We found that the major role in organelles was mitochondria by transmission electron microscope from the perspective of cell ultrastructure.
