ObjiectiveOn the basis of establishing animal model of selectivity damaged Spiral ganglion neurons(SGNs) to investigate the protective effect of Neurotrophin-3(NT-3) on the SGNs treated with ouabain, to investigate the protection mechanisms of NT-3to nerve.MethodsThe bilateral round window membranes of normal hearing mongolian gerbilses were exposed and injected80umol/L ouabain by surgery to build animal models. Neurotrophic factors-3(15ug/ml) was injected in bilateral round window membranes of successful animal models. After7days, Mongolian gerbils were sacrificed, removed the cochlea and observed the change of number and The ultrastructure of the type Ⅰ spiral ganglion cell by light microscopy and electron microscopy. The results were counted with manual counting method and analyzed with statistically by SPSS17.0.ResultsBy ouabain established animal models, the results showed that oubain was quite significantly damage the spiral ganglion cells Ⅰ, and ouabain mainly damaged the Myelin membrane surrounding and mitochondria,as well as cell counts were obviously decreased (P<0.05); For spiral ganglion cells Ⅰ, neurotrophin-3had a protective effect on the Myelin membrane surrounding, mitochondria and cell nucleus in protection group, and the cell counts is higher than injury group (P<0.05). ConclusionsNT-3can protect the type Ⅰ spiral ganglion cells which have been damaged by ouabain and reduce the injury cells., It has important clinical value for seek route of administration and effective drugs to protect cochlear nerve cells.The value of the experiment is to seek route of administration and effective drugs to protect cochlear nerve cells in clinical...
