Objective:This study was to investigate the protective effect of gastrodin (GAS) in the hydrogen peroxide (H2O2)-stimulated PC12cells.Methods: The apoptosis cell model was structured by H2O2-stimulated PC12cells, the cells were treated with GAS (80,120and160μmol/ml)prior to H2O2(200μmol/L) exposure. The effects of GAS on viability of PC12cells were reflected by the expressions of tumor necrosis factor (TNF-α)、interleukin-1P (IL-1β)、catalase(CAT) and glucocorticoid receptor(GR),which were tested by double-immunofluorescence labeling assay and reverse transcription polymerase chain reaction (RT-PCR), With Hoechst33342/propidium iodide (PI) double staining of apoptotic cells was observed morphological changes.Results: PC12cells with5x105cells/ml were seeded in six-well plates, then pretreated with80,120,160μmol/ml of GAS when the cells are grown to80%. After1h of the pretreatment, we stimulated PC12cells with H2O2(200μmol/L).Then we used double labeling immunofluorescence detection methods, RT-PCR methods, Hoechst33342/PI double staining analysis to investigate the PC12cells, and the results is: Different concentrations of GAS significantly inhibited H2O2-induced TNF-α and IL-1β secretions and activated the expression ofCAT, GR at the transcriptional and translational levels, and the degree of expression is in a dose-dependent manner.Conclusion:GAS can inhibit the release of H2O2-induced apoptosis factors of PC12cells inPI3K/Akt dependent pathway and have the anti-apoptotic potential, which have played a role in protecting neurons, suggesting GAS as an anti-apoptotic drugs of the nerve system.We haveconfirmed the potential of GAS as a anti-apoptotic drug, and have provided a new biological basis for the clinical application of GAS treatment of neurodegenerative diseases.
