| Objective:To observe sesamin collaborative vinorelbine effect on human lung adenocarcinoma cell line A-549,for providing new ideas in the chemotherapy of non-small cell lung cancer and theoretical and experimental basis of future clinical applications.Methods:Selection of human lung adenocarcinoma cell line A-549in experimental cells, and cultured in vitro. MTT was used to determine the anti-proliferative effect in human lung adenocarcinoma cell line A-549by sesamin, vinorelbine and sesamin collaborative vinorelbine;Application of inverted microscope, HE stain, Hoechst33258fluorescence microscope to observe the morphological changes of human lung adenocarcinoma cell line a-549cell apoptosis with sesamin, vinorelbine and sesamin collaborative vinorelbine, By flow cytometry. the apoptosis of human lung adenocarcinoma cell line A-549and the influence of the cell cycle were detected with vinorelbine,sesamin and combined use of two drugs; Enzyme mark is used to test the activity changes of caspase3in control group and experimental group (group sesamin, group vinorelbine, group sesamin collaborative vinorelbine).Results:①MTT results showed:sesamin concentration in the range of10μg/ml~100μg/ml,acting on human lung adenocarcinoma cell line A-549with48hours,we found that low concentrations of sesamin has an anti-proliferative effect in human lung adenocarcinoma cell line A-549, vinorelbine concentration in the range of5μg/ml~35μg/ml,acting on human lung adenocarcinoma cell line A-549with48hours, found that it can inhibit the role of human lung adenocarcinoma cell line A-549, and with the increase of drug concentration the increased of cell inhibition rate, sesamin collaborative vinorelbine in human lung adenocarcinoma cell line a-549,the growth inhibition rate is higher than two single drug (P<0.01);②Oservation of cell morphology by Inverted microscope and HE staining, Hoechst33258fluorescence staining,found that sesamin group (S40μg/ml, vinorelbine group (N20μg/ml) and the combined treatment group (S40μg/ml+N20μg/ml) acting on the human lung adenocarcinoma cell line A-549with48hours, the number of adherent cells decreased significantly, with loose connections between cells, diminished adherent capacity, some A-549human lung adenocarcinoma cells were not firmly adherent, smaller size, circular or irregular shape, nuclear chromatin condensation, membrane blebbing formation of apoptotic bodies, tumor cells lose their polygonal or spindle-shaped morphology, and can be seen more obviously in the combination group;③According to the results of Flow cytometry detection of human lung adenocarcinoma cell line A-549apoptosis rate, with48hours, sesamin group (S40μg/ml), vinorelbine group (N20μg/ml) and the combined treatment group (S40μg/mll+N20μg/ml) have a high early apoptotic rate, and in combination group,the early apoptosis rate was significantly higher than the single groups; ④Sesamin concentration in IC50(40μg/mll) to deal with human lung adenocarcinoma cell line A-549with48h. the percentage of cells decreased in GO/G1phase, increase in S-phase, cycle arrest at the S phase. Vinorelbine concentration IC50(20μg/ml) dealing with human lung adenocarcinoma cell line A-549with48h, the percentage of cells decreased in GO/G1phase, increase in G2/M phase, cycle arrest at the G2/M phase. sesamin collaborative vinorelbine were used in human lung adenocarcinoma cell line A-549with48h, the percentage of cells decreased in GO/G1phase, increase in S and G2/M phase,major in S period,so cycle arrest at the S period.⑤sesamin and vinorelbine combined effect in human lung adenocarcinoma cell line A-549, the activity of caspase-3significantly increased compared to single groups (P<0.01).Conclusion:1. Low concentration of sesamin has certain anti-proliferative effect on human lung adenocarcinoma cell line A-549;2. Sesamin combined vinorelbine have a significant anti-proliferative effect on human lung adenocarcinoma cell line A-549and can anti-tumor cell;3. From the perspectives of morphology, sesamin and vinorelbine have an induced effect on human lung adenocarcinoma cell line A-549, and the combination is more significant;4. Sesamin combined vinorelbine have a synergistic effect at inhibition of human lung adenocarcinoma A-549cells,and can reduce the dosage of vinorelbine compared to combination therapy;5. Sesamin combined vinorelbine major inhibit cell cycle S period,play a role of anti-tumor;6. Sesamin collaborative vinorelbine acting on the human lung cancer cell line A-549can increase the degree of activation of caspase-3. |
