Objective1To study the relationship of osteopontin(OPN) and bone mineral density(BMD), bone turnover markers in postmenopausal women.2To explore the effect of kidney-reinforcing, spleen-strengthening and removing blood stasis herbs on serum OPN and OPN mRNA of femoral marrow in ovariectomized rats.Methods1BMD was measured by dual-energy x-ray absorptiometry in lumbar spine1-4and left femoral neck. Levels of serum procollagen type I N-terminal propeptide (PINP), β-crosslaps (β-CTX),25-hydroxyvitamin D (250HD), parathyroid hormone (PTH), OPN, bone glaprotein (BGP), calcium (Ca) and phosphorus (P) were assessed in125postmenopausal women.2Forty female SD rats were randomly divided into four groups: sham-operated group, ovariectomized group, herbal group and alendronate group. Osteoporosis model was established by ovary resection of female rats. After treatment for12weeks, we measured bone mineral density (BMD) by small animals specially dual-energy x-ray absorptiometry in left femur. Serum OPN levels were assessed by ELISA, and femoral marrow tissue was collected to detect OPN mRNA by Quantitative PCR.Results1①Osteopontin levels were significantly higher in osteoporosis group versus osteopenic and normal group(F=0.118, P=0.000).②BMD and Ca were negative correlated with OPN, while age, β-CTX and BGP were positive (all P<0.05).③Multiple regression analysis showed that BMD of left femoral neck and Ca were independent predictors of serum OPN levels.2①Compared with the sham-operated group, BMD in ovariectomized group was significantly lower(P<0.001). Besides, it had a significant rise of BMD in herbal group and alendronate group when compared with ovariectomized group.②Serum OPN and the expression of OPN mRNA levels in ovariectomized group were significantly higher than that in sham-operated group(P<0.05).③Compared with ovariectomized group, serum OPN levels gone down slightly (P=0.049) and the expression of OPN mRNA in femoral marrow tissue decreased significantly(P<0.001) in herbal group. However, both serum OPN and the expression of OPN mRNA reduced significantly in alendronate group (P=0.001; P<0.001).④There were significant differences of serum OPN and the expression of OPN mRNA levels between herbal group and alendronate group(P=0.000; P=0.014).ConclusionOPN is very important for the occurrence and development of osteoporosis. The clinical research shows that high levels of OPN are associated with low BMD, increased levels of β-Crosslaps and the lack of Ca, and the animal experimental results show Kidney-reinforcing, spleen-strengthening and removing blood stasis herbs can restrain the absorption of bone by decreasing OPN levels, which may be one of the possible target to treat osteoporosis. They provide a new idea for controlling postmenopausal osteoporosis.
