【Objective】To observe the effect of rhaponticum uniflorum on preventingand curing mouse of Non-alcoholic fatty liver disease, and make a preliminarydiscussion on its mechanism.【Methods】72healthy SPF Kunming mice(weight18-22g)were randomlydivided into6groups, including the normal control group, the model group, thekezhijiaonang group, the rhaponticum uniflorum high dose group, therhaponticum uniflorum middle dose group, the rhaponticum uniflorum low dosegroup,12mice in each group. All of the mice were fed7days with normal SPFforage to adapt the environment. The normal control group were fed with normalSPF forage, and the other groups were fed with high fat forage, for6weeks.Allgroups of mice were given forage equal in quality daily during the period offeeding, the normal drinking water,weigh-ins weekly, in addition to the normalcontrol group, the other groups of mice were subcutaneous injection of5%CCl4cooking oil solution in volumetric fraction in hind limbs with the dose of0.1ml per10grams, twice per week.At the same time of molding in mice, the kezhijiaonang group, therhaponticum uniflorum high dose group, the rhaponticum uniflorum middle dosegroup and the rhaponticum uniflorum low dose group were given a gavage withcorresponding drug. The rhaponticum uniflorum low dose group were given agavage at2.0g·kg1every day(was10times of the clinical dosage in humans),the rhaponticum uniflorum middle dose group were given a gavage at4.0g·kg1every day(was20times of the clinical dosage in humans), the rhaponticumuniflorum high dose group were given a gavage at8.0g·kg1every day(was40times of the clinical dosage in humans), the kezhijiaonang group were given agavage at0.49g·kg1every day. The normal control group and the model groupwere given a gavage with equivalent distilled water, all the mice take food andwater freely in experimental period. At the end of the sixth week, kill all themice after a12-hour of fasting after final dose, and calculate the liver index,then the level of triglyceride, total cholesterol, alanine aminotransferase, aspartate aminotransferase, high-density lipoprotein in mice serum, the level ofmalondialdehyde, free fatty acid, glutathione and superoxide dismutase in micehepatic tissue. Observe the degree of fatty change of the mice hepatic tissueunder the optical microscope.【Results】(1)Influence on TG of mice serum with rhaponticum uniflorumdecoction: Compared with the model group, the TG of the normal control group,the kezhijiaonang group, the middle dose group and the high dose group has aobvious drop (P<0.05);compared with the normal control group, thekezhijiaonang group, the low dose group, the middle dose group and the highdose group with no statistical difference(P>0.05).(2)Influence on TC of mice serum with rhaponticum uniflorum decoction:Compared with the model group, the decrease of TC of the normal control group,the kezhijiaonang group, the low dose group, the middle dose group and the highdose group has significant statistical difference(P<0.05); compared with thenormal control group, the kezhijiaonang group, the low dose group, the middledose group and the high dose group has no statistical difference(P>0.05).(3)Influence on HDL of mice serum with rhaponticum uniflorum decoction:Compared with the model group, the increase of HDL of the low dose group,the middle dose group and the high dose group has significant statisticaldifference(P<0.05); compared with the normal control group, the kezhijiaonanggroup and each dose rhaponticum uniflorum group has no difference(P>0.05).(4)Influence on ALT of mice serum with rhaponticum uniflorum decoction:Compared with the model group, the ALT of the normal control group, thekezhijiaonang group, the low dose group, the middle dose group and the highdose group was decreased and has significant statistical difference(P<0.05);compared with the normal control group, the middle dose group and the highdose group has no statistical difference(P>0.05).(5)Influence on AST of mice serum with rhaponticum uniflorum decoction:Compared with the model group, the decrease of each dose rhaponticumuniflorum group has significant statistical difference(P<0.05);compared withthe normal control group, all the other groups has significant statisticaldifference(P<0.05).(6)Influence on MDA of mice hepatical tissues with rhaponticum uniflorumdecoction: Compared with the model group, the decrease of MDA of each dose rhaponticum uniflorum group has significant statistical difference(P<0.05);compared with the normal control group, each dose rhaponticum uniflorumgroup has no difference(P>0.05).(7)Influence on FFA of mice hepatical tissues with rhaponticum uniflorumdecoction: Compared with the model group, the decrease of FFA of the middledose group and the high dose group has difference(P<0.05); compared with thenormal control group, the kezhijiaonang group and each dose rhaponticumuniflorum group has no difference(P>0.05).(8)Influence on GSH of mice hepatical tissues with rhaponticum uniflorumdecoction: Compared with the model group, the increase of GSH of thekezhijiaonang group and each dose rhaponticum uniflorum group has significantstatistical difference (P<0.05); compared with the normal control group, thekezhijiaonang group and the low dose group has difference(P<0.05),the middledose group and the high dose group has no difference(P>0.05).(9)Influence on SOD of mice hepatical tissues with rhaponticum uniflorumdecoction: Compared with the model group, the increase of SOD of thekezhijiaonang group and each dose rhaponticum uniflorum group has significantstatistical difference(P<0.05); compared with the normal control group, eachdose rhaponticum uniflorum group and the kezhijiaonang group has nodifference(P>0.05).(10)Influence on the liver index of mice with rhaponticum uniflorumdecoction: Compared with the model group, the decrease of liver index of eachdose rhaponticum uniflorum group and the kezhijiaonang group has nodifference(P<0.05); compared with the normal control group, the liver indexof the other groups has difference(P>0.05).(11) Influence on pathomorphology of hepatic tissues of mice withrhaponticum uniflorum decoction: The liver cells of the model group wereswelling with disorder arrangement, the cytoplasm was loosen and appears meshstructure, translucence, has fat cavitation and ballooning degeneration, somecircular dark red uniform chromatoid acidophilic body or councilman body andpunctate necrosis. The damage of hepatic tissues of the drug group wasdecreased, the middle dose group and the high dose group were better than thelow dose group and the kezhijiaonang group.【Conclusion】(1)The rhaponticum uniflorum decoction can obviously improve the serum and hepatical tissue indexes of NAFLD mice;(2)Influence on the indexes of NAFLD mice with rhaponticum uniflorumdecoction were incomplete directly proportional to the dose.(3)The rhaponticum uniflorum decoction improve the injury of liver cells ofNAFLD mice may be related to its antioxidant effect.
